Dynamics of predominant microbiota in the human gastrointestinal tract and change in luminal enzymes and immunoglobulin profile during high-altitude adaptation

High-altitude (HA) visitors like pilgrims, trackers, scientists and military personnel face a group of nonspecific gastrointestinal (GI) symptoms during acclimatization to hypobaric hypoxia. In order to investigate the alteration of indigenous gastrointestinal microbiota in the development of such GI symptoms, an experiment was conducted for the enumeration of dominant cultivable faecal microbiota of 15 soldiers at base level (Delhi) and during their 15-day acclimatization at 3,505 m HA (Leh). At HA, faecal microbiota analysis revealed that total aerobes decreased significantly with increase of total and facultative anaerobes. The strict anaerobes like Bifidobacterium sp., Bacteroidetes sp. and Lactobacillus sp. exhibited positive growth direction index (GDI) like other predominant obligate anaerobes Clostridium perfringens and Peptostreptococcus sp. Different enzymes like amylase, proteinase and polyphenol hydrolase produced by different bacterial populations showed positive GDI, whereas phosphatase producers exhibited negative GDI. The levels of microbe-originated enzymes like amylase, proteinase, alkaline phosphatase and beta-glucuronidase were also elevated during HA acclimatization. In addition, in vitro gas production ability was enhanced with increase of faecal immunoglobulins IgA and IgG. We demonstrated that hypoxic environment at HA had the potential to alter the gut microbial composition and its activities that may cause GI dysfunctions.

Production of glutathione from probiotic Bacillus amyloliquefaciens KMH10 using banana peel extract

Summary: The aim of this study was to isolate a highly beneficial probiotic bacterium capable of producing glutathione. The isolated strain, Bacillus amyloliquefaciens KMH10, exhibited antioxidative activity (77.7 ± 2.56) and several other essential probiotic attributes. A consortium of lignocellulolytic enzymes was used to saccharify banana peel, resulting in the production of 65.71 g/L sugar to support optimal glutathione production of 181 ± 4.56 mg/L, which was 1.6 times higher than the control. Therefore, this probiotic bacteria could serve as an effective resource for glutathione and be used as natural therapeutics for the prevention/treatment of inflammation-related gastric ailments, as well as an effective producer of glutathione using valorized banana waste that has excellent industrial relevance.

Attenuation of fluoride-induced hepatorenal oxidative stress by ferulic acid in vivo: An approach with in-silico analysis and interaction informatics of ferulic acid

Summary: This study aimed to investigate the defensive impact of ferulic acid against sodium fluoride-induced hepatorenal dysfunction at the biochemical and antioxidative systems. Ferulic acid remarkably defended against oxidative damage, reduced lipid peroxidation, and normalized the serum biomarkers. In-silico and in-vitro experiments further validated the protective effects of ferulic acid on the activity of antioxidant enzymes.

Customized molecular diagnostics of bacterial bloodstream infections for carbapenem resistance: A convenient and affordable approach

Summary: The acute crisis of carbapenem resistance hinders the use of carbapenems in medical emergencies, especially bloodstream infections. A low-cost in-house molecular diagnostics assay was developed to rapidly detect carbapenemase-producing carbapenem-resistant organisms (CP-CROs) in positive blood-culture broths. The assay has been validated and meets the minimal requirements of the WHO for multiplex-PCR.

Extraction of chitosan from biologically-derived chitin by bacterial chitin deacetylase: Process optimization and product quality assessment

Summary: This study focuses on the enzymatic extraction of chitosan from chitin, avoiding the adverse effects of chemical treatment and reducing environmental pollution. A bacterial strain, Alcaligens faecalis CS4, was identified as a potent chitin deacetylase (CDA) producer. After optimization, a CDA production of 40.69 U/mL was achieved. The resulting chitosan exhibited desirable properties, including high solubility, degree of deacetylation, crystallinity index, molecular weight, and decomposition temperature. It also demonstrated antioxidant potential, antimicrobial activity, mucoadhesiveness, and cholesterol binding properties.

Characterization of a novel lytic bacteriophage VPMCC14 which efficiently controls Vibrio harveyi in Penaeus monodon culture

Summary: The bacteriophage VPMCC14 was characterized for its lytic ability against Vibrio harveyi, the causative agent of luminous vibriosis diseases in shrimp. VPMCC14 showed strong lytic activity against V. harveyi in liquid culture and was able to control V. harveyi infection and reduce shrimp mortality in aquariums.
