A stimulatory effect of apigenin-6-C-beta-fucopyranoside (1) on glucose uptake was observed when rat soleus muscle was incubated with 1, 10 and 100 mu M of this flavonoid glycoside. The presence of specific insulin signaling inhibitors, such as wortmannin, an inhibitor of phosphoinositide 3-kinase (P13K), RO318220, an inhibitor of protein kinase C (PKC), PD98059, an inhibitor of mitogen-activated protein kinase (MEK), and HNMPA(AM)(3), an insulin receptor tyrosine kinase activity inhibitor showed that apigenin-6-C-beta-fucopyranoside triggers different metabolic pathways in skeletal muscle. The oral administration of crude extract, fractions and isolated flavonoids (apigenin-6-C-beta-fucopyranoside (1) and apigenin-6-C-(2 ''-O-alpha-rhamnopyranosyl)-beta-fucopyranoside (2)) from Averrhoa carambola leaves exhibited a potential hypoglycemic activity in hyperglycemic normal rats. Additionally, both flavonoids significantly increased the muscle and liver glycogen content after an acute treatment. The results indicate that A. carambola can be regarded as a potent anti hyperglycemic agent with insulin secretagogue and insulin mimetic properties. (C) 2012 Elsevier B.V. All rights reserved.

Summary: Cardiac glycosides have potent cytotoxic and anticancer effects, with the natural cardenolide glucoevatromonoside (GEV) showing short- and long-lasting cytotoxic effects on non-small-cell lung cancer H460 cells. GEV reduced viability, migration, and invasion of H460 cells, induced cell cycle arrest and death, and inhibited Na+,K+-ATPase activity. Additionally, combining GEV with chemotherapeutic drugs exhibited synergistic antiproliferative effects, especially with cisplatin and paclitaxel. The study also revealed a link between Na+,K+-ATPase and Cl- ion channels.

Pyriproxyfen induces intracellular calcium overload and alters antioxidant defenses in Danio rerio testis that may influence ongoing spermatogenesis

Vanessa Staldoni de Oliveira, Allisson Jhonatan Gomes Castro, Katiuska Marins, Ana Karla Bittencourt Mendes, Gabriel Adan Araujo Leite, Ariane Zamoner, Glen Van der Kraak, Fatima Regina Mena Barreto Silva

Summary: The study revealed that pyriproxyfen can affect the ionic balance in the testis of zebrafish through multiple pathways, leading to oxidative stress and impacting reproductive health. Pyriproxyfen may induce toxicity in the testis by causing calcium overload, increased lipid peroxidation, and diminished antioxidant capacity, thereby affecting spermatogenesis.

Triterpene betulin may be involved in the acute effects of pulp and paper mill effluent on testis physiology in zebrafish

Summary: This study compared the effects of pulp and paper mill effluent and betulin on various aspects of testicular physiology in zebrafish, indicating that the alterations may be due to the actions of betulin. Exposure to both effluent and betulin led to decreased lactate and calcium influx, possibly through the activation of the NCX pump. Additionally, betulin treatment resulted in increased ROS, GSH content, GST activity, and a tendency towards decreased CAT activity in the testes.

Summary: The study investigated the effects of Bisphenol A (BPA) on testicular energy metabolism and morphology in zebrafish through in vitro and short-term in vivo experiments. Results showed that BPA exposure decreased lactate and glycogen content, altered enzyme activity, induced apoptosis in testicular cells, and affected spermatogenesis in male zebrafish. Overall, short-term BPA exposure was found to impact energy metabolism and spermatogenesis in zebrafish.

In vivo exposure to pyriproxyfen causes ovarian oxidative stress and inhibits follicle maturation in zebrafish

Summary: We studied the effects of pyriproxyfen on zebrafish ovarian morphology and oxidative stress. Exposure to pyriproxyfen resulted in increased generation of reactive oxygen species and decreased glutathione content in the ovaries. Additionally, enzyme activities related to antioxidative defense mechanisms were altered. The histological analysis showed disrupted follicular maturation and various abnormalities in the ovary. These findings suggest that pyriproxyfen acts as an endocrine disruptor and can impair fish reproduction.

Summary: This study evaluated the anti-diabetic activity of the butanol fraction from Physalis peruviana in induced type 2 diabetic mice. The results showed that the butanol fraction at a higher dose (100 mg/kg) had better regulation of blood glucose levels in mice, improved lipid metabolism, and protected against oxidative stress.

Signal transduction of the insulin secretion induced by the chalcone analogue, (E)-3-(phenyl)-1-(3,4,5-trimethoxyphenyl)prop-2-en-1-one, and its role in glucose and lipid metabolism

Summary: A chalcone analogue, DMU 101, was synthesized and investigated for its antidiabetic properties. It was found to decrease blood glucose levels, increase glycogen content in skeletal muscle, and reduce serum triacylglycerol and total cholesterol levels. Furthermore, the analogue stimulated glucose uptake and increased insulin secretion through calcium-dependent mechanisms. These findings suggest that this chalcone may have potential for treating diabetes by regulating glucose and lipid metabolism.

Dibutyl phthalate disrupts energy metabolism and morphology in the gills and induces hepatotoxicity in zebrafish

Keyla Rodrigues, Hemily Batista-Silva, Kieiv Resende Sousa de Moura, Glen Van Der Kraak, Fatima Regina Mena Barreto Silva

Summary: This study examined the immediate impacts of dibutyl phthalate (DBP) exposure on energy metabolism and gill histology in zebrafish. In vitro exposure to 10 μM DBP for 60 minutes resulted in altered energy supply in the gill tissue, with decreased lactate content and lactate dehydrogenase (LDH) activity. Higher DBP concentrations (100 μM and 1 mM) increased lactate content and LDH activity, blocked glucose uptake, depleted glycogen content, and caused gill injury and liver damage.
