Summary: Using time-lapse embryo staging and single-cell sequencing, Meistermann et al. (2021) provide new insights into the precise timing of lineage specification during human preimplantation development and raise questions about the concept of a human inner cell mass.

Summary: This review discusses the sources of DNA damage in gametes and preimplantation embryos, with a focus on current knowledge, limits in DNA damage detection, and screening strategies. It also explores obstacles and future perspectives in the clinical diagnosis and treatment of DNA damaged embryos.

Summary: MicroRNAs play a crucial role in preimplantation embryo development, with their expression increasing during the morula and blastocyst stages. Knockdown of DGCR8 inhibits the expression of embryonically expressed miRNAs and the morula-to-blastocyst transition.

Summary: Recent evidence from time-lapse technology and preimplantation genetic testing highlights the remarkable plasticity of human embryos in vitro, challenging current embryological notions and suggesting the need for new rules governing preimplantation development.

Summary: This study investigates the use of a novel zinc chelator for activating bovine and porcine oocytes. The results show that the zinc chelator can successfully induce blastocyst development in both species, although developmental rates and embryo quality are compromised in bovine compared to calcium-induced embryos. On the contrary, the new concentration and incubation time of the zinc chelator improve developmental rates in porcine without affecting embryo quality. These findings contribute to understanding the role of zinc in oocyte activation and preimplantation embryo development across different mammalian species.

Summary: This study evaluated the efficiency of in vitro embryo splitting procedures and assessed the quality of blastocysts from different embryo sources. Chromosomally normal embryos were found to be more likely to develop into viable blastocysts. Despite poor blastocyst quality, 1PN and 3PN embryos were identified as the best options for splitting procedures in research contexts.

Summary: The study reveals that glutamic acid can affect preimplantation embryo development through cell membrane receptors. High extracellular concentrations of glutamic acid can inhibit blastocyst development, which is of practical significance.

Summary: DKK1 can alter the early developmental program of bovine embryos, affecting the number of trophectoderm cells and placental function. Treatment with DKK1 at the morula-to-blastocyst stages has long-term consequences on postnatal phenotype, resulting in heavier calves at birth but slower growth after birth.

Summary: During in-vitro maturation of immature porcine oocytes, gonadotropins are not necessary when FGF2, LIF, and IGF1 are present. FLI directly impacts oocyte competence without the need for gonadotropins, and in the presence of gonadotropins, it enhances cumulus cell expansion.

Summary: The study showed that VEGF-loaded silica supraparticles (V-SPs) can promote embryonic development in vitro, similar to the effect of free VEGF supplementation. This suggests that V-SPs could be a novel method for delivering therapeutic agents to support embryo development, potentially overcoming the limitations of VEGF stability in solution. Further investigation is needed to explore the clinical application of V-SPs in IVF.

Summary: This study compared the developmental competence of oocytes that matured in vivo to metaphase II (MII) with those that matured in vitro from metaphase I (MI). The results showed that MII oocytes had higher fertilization rates, blastocyst formation rates, and euploidy rates compared with MI-MII oocytes. However, there was no significant difference in clinical pregnancy rates, spontaneous pregnancy loss rates, and live birth rates between the two groups after single euploid blastocyst transfers.

Summary: This study found that corticotropin-releasing hormone (CRH) and cortisol indirectly impair pig fertilization and embryo development by inducing apoptosis in pig oviductal epithelial cells (OECs) through the activation of the FAS and TNF alpha systems. Adrenocorticotropic hormone (ACTH) did not show any detrimental effect on pig embryos or OECs.
