In rat experimental varicocele, polydeoxyribonucleotide (PDRN) induces vascular endothelial growth factor (VEGF) production, thereby enhancing testicular function. This may point to a new therapeutic approach in human varicocele. (Fertil Steril (R) 2011;95:1510-3. (C) 2011 by American Society for Reproductive Medicine.)

CXCL12/CXCR4 axis supports mitochondrial trafficking in tumor myeloma microenvironment

Cesarina Giallongo, Ilaria Dulcamare, Daniele Tibullo, Vittorio Del Fabro, Nunzio Vicario, Nunziatina Parrinello, Alessandra Romano, Grazia Scandura, Giacomo Lazzarino, Concetta Conticello, Giovanni Li Volti, Angela Maria Amorini, Giuseppe Musumeci, Michelino Di Rosa, Francesca Polito, Rosaria Oteri, M'hammed Aguennouz, Rosalba Parenti, Francesco Di Raimondo, Giuseppe A. Palumbo

Summary: In multiple myeloma, mesenchymal stromal cells (MSCs) promote tumor growth and provide metabolic support to plasma cells (PCs) through mitochondrial transfer. Myeloma MSCs rely less on mitochondrial metabolism compared to healthy MSCs and are more prone to delivering mitochondria to myeloma cells. Additionally, PCs induce the expression of connexin 43 (CX43) in MSCs, leading to the activation of CXCL12 and the stimulation of its receptor CXCR4 on myeloma cells, promoting mitochondrial transfer. These findings suggest that the CXCL12/CXCR4 axis mediates intercellular communication in the myeloma niche.

Summary: The study found that there are complex epigenetic modifications, mainly methylation and miRNA activity, in pituitary adenomas. Nonfunctioning tumors (NFPAs) showed higher methylation levels compared to GH-omas, with 178 differentially methylated regions (DMRs) mainly located in noncoding and intronic sequences. Additionally, the study identified three hypermethylated genes involved in tumorigenesis processes.

Nutraceuticals as Alternative Approach against Cadmium-Induced Kidney Damage: A Narrative Review

Herbert Ryan Marini, Federica Bellone, Antonino Catalano, Giovanni Squadrito, Antonio Micali, Domenico Puzzolo, Jose Freni, Giovanni Pallio, Letteria Minutoli

Summary: Cadmium (Cd) poses a risk to public health due to its non-biodegradability and long half-life in the body. Cd mainly targets the kidneys and accumulates there. Studies have shown that Cd exposure can lead to bone fragility, not only through direct toxic effects on bone mineralization but also due to renal failure. Further research has revealed the molecular pathways through which Cd induces serious kidney damage, including lipid peroxidation, inflammation, programmed cell death, and hormonal imbalances. Chronic kidney disease (CKD) is associated with dysbiosis, and recent studies have highlighted the altered composition and functions of gut microbiota in CKD. Considering the strong connection between diet and CKD management, nutraceuticals found in Mediterranean diet can be a safe therapeutic strategy for Cd-induced kidney damage and contribute to the prevention and treatment of CKD.

Glutathione: Lights and Shadows in Cancer Patients

Herbert Ryan Marini, Bianca Arianna Facchini, Raffaele di Francia, Jose Freni, Domenico Puzzolo, Liliana Montella, Gaetano Facchini, Alessandro Ottaiano, Massimiliano Berretta, Letteria Minutoli

Summary: In cases of cellular injury, excessive production of reactive oxygen species (ROS) can lead to various conditions, including cancerogenesis. Glutathione (GSH), the most abundant thiol-containing antioxidant, is crucial for restoring redox homeostasis. The conjugation of GSH with xenobiotics, such as anti-cancer drugs, can either reduce their harmful effects or enhance their toxicity. Further studies are needed to better understand the relationship between GSH and cancer. The use of GSH should only be considered under medical supervision, taking into account the appropriate timing and setting, as self-prescribed GSH lacks strong scientific evidence for its efficacy in reducing toxicity.

Biglycan Involvement in Heart Fibrosis: Modulation of Adenosine 2A Receptor Improves Damage in Immortalized Cardiac Fibroblasts

Michele Scuruchi, Federica Mannino, Chiara Imbesi, Giovanni Pallio, Giovanna Vermiglio, Gianluca Bagnato, Letteria Minutoli, Alessandra Bitto, Francesco Squadrito, Natasha Irrera

Summary: Cardiac fibrosis is characterized by abnormal deposition of extracellular matrix proteins, myofibroblast differentiation, and collagen deposition stimulated by TGF-beta activation. Biglycan (BGN) plays a key role in matrix assembly and the control of cardiac fibroblast phenotype. Adenosine receptors (ARs), particularly A(2AR), may stimulate fibrotic damage through collagen production, and A(2AR) modulation could be an effective therapeutic option to reduce fibrotic processes in heart remodeling.
