Transient receptor potential vanilloid-1 signaling inhibits differentiation and activation of human dendritic cells

The goal of the current study was to investigate the expression of transient receptor potential vanilloid-1 (TRPV1) on human in vitro differentiated monocyte-derived dendritic cells (DCs) and to dissect the corresponding role of TRPV1-signaling in DC-specific functions. TRPV1 expression was identified both at the protein and gene levels in human DCs. Moreover, the prototypic TRPV1 agonist capsaicin specifically (i.e. via TRPV1) and dose-dependently inhibited cytokine-induced DC differentiation, phagocytosis of bacteria, activation of DCs, and pro-inflammatory cytokine secretion. These data introduce TRPV1-coupled signaling as a novel player in human monocyte-derived DC biology with anti-inflammatory actions. (C) 2009 Federation of European Biochemical Societies. Pruritus: A Sensory Symptom Generated in Cutaneous Immuno-Neuronal Crosstalk

Summary: Pruritus, or itch, is a common symptom associated with various skin and immune disorders. Despite significant progress in understanding the molecular mechanisms behind acute and chronic itch, there are still gaps in our knowledge, making the clinical management of pruritic conditions challenging. Recent research has highlighted the interactions between pruriceptive sensory neurons and different cutaneous cell types, particularly in inflammatory conditions, where these cells release mediators that excite the itch-sensing fibers. There is also bidirectional communication, with sensory neurons releasing mediators that maintain an inflamed and itchy tissue environment.

TRPV4 Activation Increases the Expression of CD207 (Langerin) of Monocyte-Derived Langerhans Cells without Affecting their Maturation

Summary: This study compared the transcriptome of monocyte-derived Langerhans cells (moLCs) with that of immature dendritic cells using RNA sequencing. The results showed that moLCs express genes characteristic of LC2 cells and TRPV4. Activation of TRPV4 increased the expression of Langerin and intracellular calcium concentration in moLCs. Furthermore, TRPV4 activation mitigated the inflammatory responses of moLCs by decreasing cytokine production and T-cell activating capability.

Identification of histidine-Ni (II) metal complex by Raman spectroscopy

Summary: Metal ions, like nickel, are strongly bound to amino acids and form metal complexes with different geometric structures. The formation of histidine-nickel (II) complex was examined under different conditions using Raman spectroscopy. Additionally, density functional theory calculations were performed to understand the formation mechanism and optimal geometry of the complex. The Raman measurements showed specific peaks that belong to the metal complex. The alkaline pH favored complex formation, while the concentration of metal ions had no effect on the formation of the histidine-nickel (II) complex.
