A splice variant of the Neurospora crassa hex-1 transcript, which encodes the major protein of the Woronin body, is modulated by extracellular phosphate and pH changes

The Woronin body, a septal pore-associated organelle specific to filamentous ascomycetes, is crucial for preventing cytoplasmic bleeding after hyphal injury. In this study, we show that T1hex-1 transcript and a variant splicing T2hex-1 transcript are up-regulated at alkaline pH. We also show that both hex-1 transcripts are overexpressed in the preg(c), nuc-1(RIP), and pacC(ko) mutant strains of Neurospora crassa grown under conditions of phosphate shortage at alkaline pH, suggesting that hex-1 transcription may be coregulated by these genes. In addition, we present evidence that N. crassa PacC also has metabolic functions at acidic pH. (C) 2008 Federation of European Biochemical Societies. Published by Elsevier B. V. Structural and functional insights of the catalytic GH5 and Calx-β domains from the metagenome-derived endoglucanase CelE2

Summary: This study evaluated the structural and functional properties of the individualized GH5 and Calx-beta domains of CelE2 by subcloning and heterologously expressing them in E. coli. The results showed that the denaturation temperature of the catalytic domain (CelE21-381) decreased compared to the full-length CelE2, and the Calx-beta domain (CelE2382-477) was unfolded without attachment to the catalytic core. The deletion of the Calx-beta domain increased the activity of the catalytic domain towards the insoluble substrate Avicel, without altering other important functional properties.

This review discusses various biochemical pathways for the biocatalytic conversion of hydroxycinnamic acids ferulic, caffeic, and p-coumaric acid into high-value molecules in the development of a biorefinery concept.

Addressing Microbial Resistance Worldwide: Challenges over Controlling Life-Threatening Fungal Infections

Summary: Fungal infections, especially those caused by drug-resistant fungi, pose a serious global health concern. Candida, Aspergillus, and Cryptococcus are some of the fungi that cause severe and systemic infections, with high mortality rates. Understanding the immunopathogenesis of fungal diseases and finding ways to prevent dissemination and deaths caused by fungal infections are crucial. This review highlights the importance of drug resistance in clinically significant fungi and discusses strategies to combat these infections.

Transcriptome Analysis of Co-Cultures of THP-1 Human Macrophages with Inactivated Germinated Trichophyton rubrum Conidia

Summary: This study analyzed the transcriptome of THP-1 cells co-cultured with inactivated germinated T. rubrum conidia to characterize deep infection. Results showed increased release of IL-12 during co-culture, while the other cytokines remained unchanged. Gene expression analysis identified 83 modulated genes involved in signal transduction, cell communication, and immune response pathways. The study revealed the ability of macrophages to modulate the immune response and release proinflammatory cytokines, providing clues for exploring molecular targets in antifungal therapies involving immune system activation.

This research expands our understanding of ligninolytic metabolism in a non-conventional yeast and provides important clues for lignin valorization in other systems.
