Evaluation of Cell Models to Study Monocyte Functions in PMM2 Congenital Disorders of Glycosylation

Summary: Congenital disorders of glycosylation (CDG) are a group of genetic metabolic diseases characterized by abnormal synthesis, attachment, or processing of protein glycosylation. Recent studies have shown immunological dysfunctions in some types of CDG. However, there is still limited research on immune cell dysfunction in PMM2-CDG and other CDG types. Immune cell models that mimic PMM2-CDG could overcome the challenges of studying patient immune cells, such as limited availability of patient material. Initial observations show abnormal surface glycosylation patterns and slightly increased uptake of fungal particles in PMM2-CDG monocytes. The use of PMM2 knockdown THP-1 monocytes as a model may be superior to tunicamycin-treated THP-1 cells for studying immune dysfunction in CDG.

Summary: Protein tyrosine phosphatases and protein tyrosine kinases play important roles in controlling molecular signaling processes at cellular and organismal levels, affecting the quality of life. This review focuses on studying germline variants in the genes encoding the thirty-seven classical members of the protein tyrosine phosphatase superfamily. The results suggest a potential association between individual genes and specific disorders, highlighting the need for further research to establish a comprehensive genotype-phenotype map for this intriguing protein family.

CYSRT1: An Antimicrobial Epidermal Protein that Can Interact with Late Cornified Envelope Proteins

Summary: Late cornified envelope (LCE) proteins are small cationic epidermal proteins with antimicrobial properties. We identified CYSRT1 as an interacting partner of LCE proteins, which is specifically expressed in cutaneous and oral epithelia and colocalizes with LCE proteins in the upper layers of the suprabasal epidermis. CYSRT1 has antimicrobial activity against Pseudomonas aeruginosa and contributes to the constitutive epidermal antimicrobial host defense repertoire.
