Summary: Research shows that the loss of Flt4 specifically in lymphatic cells inhibits the formation of specialized button junctions in multiple tissues and impairs interstitial absorption. Knockdown of FLT4 in human lymphatic endothelial cells results in disrupted NOTCH1 expression and activation, while overexpression of the NOTCH1 intracellular domain in Flt4 knockout vessels rescues the formation of button junctions and absorption of interstitial molecules. These findings reveal the importance of VEGFR3 and NOTCH1 signaling in the development of button junctions during postnatal development and may have clinical implications for lymphatic diseases with impaired VEGFR3 signaling.

Summary: In this study, the dynamics and molecular mechanisms of lymphatic endothelial cell (LEC) renewal in adult mesenteric lymphatic vessels were investigated. It was found that adult LEC turnover and proliferation were limited to the valve regions of collecting vessels, with valve cells having the shortest lifespan. Proliferating valve sinus LECs were identified as the main source for maintenance and repair of lymphatic valves. Mechanistic target of rapamycin complex 1 (mTORC1) was identified as a mechanoresponsive pathway activated by fluid shear stress in LECs, driving valve cell division or dictating their mechanic resilience through increased protein synthesis. Overactivation of lymphatic mTORC1 in vivo promoted supernumerary valve formation.

Summary: Secondary lymphedema is a debilitating disease caused by dysfunction of the lymphatic system, characterized by abnormal soft tissue swelling. Despite its high prevalence, there are no approved therapeutic agents that can prevent disease progression. Previous studies have shown that 9-cis-retinoic acid (9-cisRA) has the potential to be repurposed for lymphedema by promoting lymphangiogenesis. This study demonstrates that the nuclear receptor RXR alpha plays a critical role in the therapeutic efficacy of 9-cisRA in treating postsurgical lymphedema.
