Nude mice exhibit athymia and hairlessness by a loss-of-function mutation in the transcription factor Foxn1 gene. Although the immunological functions of Foxn1 have been studied intensively, there have been relatively few studies of its functions in skin. Foxn1 regulates expression of hair keratins, which is essential for normal hair structure; however, how Foxn1 regulates hair keratin expression and hair formation is largely unknown. In the present study, we found that mice lacking phospholipase C (PLC)-delta 1, a key molecule in the phosphoinositide signaling pathway, and nude mice show similar hair abnormalities, such as lack of cuticle and bending. We also found that expression of hair keratins was remarkably decreased in skin of PLC-delta 1 knockout mice. Furthermore, expression of PLC-delta 1 was induced in Foxn1-transfected U2OS cells. In addition, we showed that PLC-delta 1 expression was remarkably decreased in skin of nude mice. In skin and keratinocytes of nude mice as well as PLC-delta 1 KO mice, activation of PLC downstream effectors, such as PKC and nuclear factor of activated T cells, was impaired. These results indicate that PLC-delta 1 is an essential molecule downstream of Foxn1 in normal hair formation, and strongly suggest that hairlessness in nude mice is caused by insufficient expression of PLC-delta 1.

Summary: The study revealed that the PLC delta 1 isoform plays a crucial role in the activation of Delta N-TRPV1 channels and osmosensory transduction in MNCs, with its absence leading to impaired MNC function. This research advances our understanding of the molecular mechanisms of osmoregulation in mammals.

PLEKHA5 regulates the survival and peritoneal dissemination of diffuse-type gastric carcinoma cells with Met gene amplification

Summary: This study identified PLEKHA5 as a protein downstream of Met that is tyrosine-phosphorylated in DGC cells with Met gene amplification. Knockdown of PLEKHA5 selectively suppressed the growth of these cells by inducing apoptosis and abrogated malignant phenotypes, suggesting its essential role in Met-addicted DGC progression. Mechanistically, PLEKHA5 dysregulation of glycolytic metabolism activates the JNK pathway to promote apoptosis.

SHP2 as a Potential Therapeutic Target in Diffuse-Type Gastric Carcinoma Addicted to Receptor Tyrosine Kinase Signaling

Summary: Diffuse-type gastric carcinoma (DGC) is characterized by aggressive progression associated with rapid infiltrative growth, massive fibrosis, and peritoneal dissemination. In this study, SHP2 was identified as a critical regulator downstream of the amplified Met and FGFR2 receptor tyrosine kinases (RTKs) in DGC cell lines. Targeting SHP2 selectively suppressed the growth and malignant phenotypes of RTK-addicted DGC, highlighting its potential as a therapeutic target for DGC treatment.

Summary: Changes in cell mechanics contribute to cancer cell dissemination. High plasma membrane (PM) tension inhibits cancer dissemination by counteracting mechanosensitive BAR family protein assembly, while restoration of PM tension phenotypically convert malignant cells into a non-motile epithelial cell state.

Transferrin receptor 1 promotes the fibroblast growth factor receptor-mediated oncogenic potential of diffused-type gastric cancer

Summary: The study reveals that in diffuse-type gastric cancer cells, transferrin receptor 1 (TfR1) binds and phosphorylates with fibroblast growth factor receptor 2 (FGFR2), promoting cell proliferation and tumorigenicity. This suggests that controlling TfR1 function could serve as a therapeutic target in diffuse-type gastric cancer with activated FGFR2.
