Baculovirus vector as a delivery vehicle for influenza vaccines

The baculovirus vector has emerged as an efficient delivery vehicle for influenza vaccines. In addition to the ease and safety in expeditious production, recent improvements in baculovirus engineering to display foreign proteins on the surface and to express transgenes with suitable promoters in various cell lines have become milestones in the development of the baculovirus expression system. Surface-displayed and shuttle promoter-mediated baculovirus vaccines for influenza present advantages in immunogenicity and safety, as studied in several animal models. A variety of strategies, including the modification of envelope proteins for surface display, the selection of novel promoters for in vivo transductions and advancements in downstream processing, aid the improvement of baculovirus-based influenza vaccines and represent progress toward next-generation vaccines for influenza.

Summary: The expression of histone deacetylase 1 (HDAC1) is reduced during classical swine fever virus (CSFV) infection, and HDAC1 acts as a negative regulator of CSFV replication by activating specific immune pathways. The N-protein of CSFV interacts with transcriptional regulator Sp1 and promotes its degradation, leading to the downregulation of HDAC1 expression. This study provides insights into the mechanisms by which CSFV evades host innate immune responses.

Nanobodies against porcine CD163 as PRRSV broad inhibitor

Summary: In this study, a phage display VHH library was constructed to select nanoantibodies against porcine CD163 receptor. Seven CD163-specific nanoantibodies were identified and showed strong affinity to CD163 receptor and excellent antiviral activity. The nanoantibodies mainly exerted antiviral functions by interfering with virus attachment stage and decreasing the transcription of CD163. The findings provide insights into the biofunction of CD163 in antiviral infection and the development of broad-spectrum strategies against PRRSV.
