Summary: Drug-induced liver injury (DILI) is a common and serious adverse drug reaction that often leads to drug development failure and withdrawal. Enhanced levels of reactive oxygen species (ROS) have been identified as a major mechanism of DILI. Human-derived cell models, such as Upcyte human hepatocytes and HepaRG cells, are suitable for long-term hepatotoxicity assessments and mechanistic studies. These cell models maintain the expression and functionality of phase I and phase II enzymes, as well as antioxidant enzymes, making them suitable for routine toxicological assessments during drug preclinical testing.

Summary: A study identified multiple viral mutations associated with resistance to tenofovir in a chronic hepatitis B patient who showed suboptimal response after 4 years of treatment. The newly discovered rtT301A mutation significantly impairs HBV replication and reduces sensitivity to tenofovir in hepatoma cell lines, but this effect is restored in primary human hepatocytes. This highlights the importance of validating antiviral drug resistance in different liver cell types to understand viral fitness under treatment pressure.

Summary: Non-alcoholic fatty liver disease (NAFLD) is characterized by lipid accumulation in hepatocytes, but the role of different macronutrients remains unclear. This study investigated the effects of fatty acids, glucose, and fructose on lipid accumulation in human hepatocytes and different cell lines. Results showed that all cell types accumulated lipid in response to fatty acids, while only primary human hepatocytes increased lipid accumulation in response to fructose. There was considerable inter-individual variability in lipid accumulation in primary human hepatocytes.

Summary: The combined exposure of arsenic and palmitic acid induced higher levels of cell toxicity, oxidative stress, lipid accumulation, and triglyceride content compared to individual exposures. Metabolomics and gene expression analysis showed that the combined effects of arsenic and palmitic acid resulted in more toxic responses in HepG2 cells.

Summary: This study used atomic force microscopy to extract the three-dimensional morphology and mechanical information of hepatocytes and hepatoma cells in culture, and compared and analyzed their characteristics. By training machine learning algorithms, the detection and classification of cells were realized with a high accuracy of 94.54% and an AUC of 0.99. This method accurately identifies and assesses hepatocytes and hepatoma cells, providing an objective basis for the early diagnosis of hepatocellular carcinoma.
