Akirins in sea lice: First steps towards a deeper understanding

Sea lice (Copepoda, Caligidae) are the most widely distributed marine pathogens in the salmon industry. Vaccination could be an environmentally friendly alternative for sea lice control; however, research on the development of such vaccines is still at an early stage of development. Recent results have suggested that subolesin/akirin/my32 are good candidate antigens for the control of arthropod infestations, including sea lice, but background knowledge about these genes in crustaceans is limited. Herein, we characterize the my32 gene/protein from two important sea lice species, Caligus rogercresseyi and Lepeophtheirus salmonis, based on cDNA sequence isolation, phylogenetic relationships, three dimensional structure prediction and expression analysis. The results show that these genes/proteins have the main characteristics of akirins from invertebrates. In addition, immunization with purified recombinant my32 from L. salmonis elicited a specific antibody response in mice and fish. These results provide an improvement to our current knowledge about my32 proteins and their potential use as vaccine candidates against sea lice in fish. (C) 2013 Elsevier Inc. All rights reserved.

Solving the enigma of POLD1 p.V295M as a potential cause of increased cancer risk

Summary: Germline variants affecting the proofreading activity of POLE and POLD1 can predispose to colorectal adenomas and carcinomas. Disruptive variants and missense variants outside the exonuclease domain (ED) of these polymerases generally do not predispose to cancer, but specific variants may indirectly affect proofreading activity. The variant POLD1 c.883G>A; p.(Val295Met) located upstream of the ED does not alter proofreading activity or associate with cancer.

Systematic Analysis of FASTK Gene Family Alterations in Cancer

Summary: The FASTK family of proteins play a crucial role in post-transcriptional regulation of mitochondrial gene expression. Genetic alterations are commonly observed in FASTK and FASTKD3, mainly consisting of amplifications, while FASTKD1 and FASTKD5 are frequently mutated. Upregulation of mRNA levels of all members of FASTK family is seen in esophageal, stomach, liver and lung cancers. The interaction of FASTK proteins with cancer signaling pathways highlights their potential as diagnostic and therapeutic targets for certain types of cancer.

Summary: COVID-19 is a highly contagious respiratory viral disease caused by the novel coronavirus SARS-CoV-2. Researchers have developed a recombinant protein by fusing the receptor-binding domain (RBD) of the viral protein spike (S) with the extracellular domain of CD154, an immune system modulator molecule. This recombinant protein has shown promising immunogenicity in mice and non-human primates, inducing a specific and high IgG response and neutralizing the virus. It has the potential to enhance the current vaccines against COVID-19.

Canonical or noncanonical? Structural plasticity of serine protease-binding loops in Kunitz-STI protease inhibitors

Summary: The Kunitz-Soybean Trypsin Inhibitor (Kunitz-STI) family is a large family of proteins with most of its members being protease inhibitors. This study analyzed the crystallographic structures of Kunitz-STI inhibitors to characterize the structural features and locations of serine protease-binding loops. The findings suggest a relationship between the conformation of these loops, the inhibition mechanism, and the plant source of the inhibitors. This research challenges the perception that all Kunitz-STI inhibitors are canonical and will aid in the identification and design of multifunctional proteins.

Recommendations for the classification of germline variants in the exonuclease domain of POLE and POLD1

Summary: Recommendations have been developed for the classification of non-disruptive variants located in the exonuclease domain of POLE and POLD1, based on the ACMG/AMP criteria. A total of 128 exonuclease domain variants reported in the literature and/or public databases were classified, with 17 variants classified as pathogenic or likely pathogenic, and 17 as benign or likely benign. These recommendations aim to improve diagnostic performance and prevent variant misclassifications in the clinical and scientific communities.

Summary: In this study, a monoclonal antibody (mAb) specific to Nile tilapia IgT was generated and confirmed for its specificity. The mAb showed immunoreactivity against recombinant IgT fragment and native IgT in skin mucus. The percentage of IgT+ B cell populations differed between juveniles and adults and after immunostimulation. This mAb serves as an important immunological tool for studying the biological function and structural characteristics of tilapia IgT.
