High glucose and oxidative/nitrosative stress conditions induce apoptosis in retinal endothelial cells by a caspase-independent pathway

Diabetic retinopathy (DR) is a leading cause of vision loss among working-age adults. Retinal endothelial cell apoptosis is an early event in DR, and oxidative stress is known to play an important role in this pathology. Recently, we found that high glucose induces apoptosis in retinal neural cells by a caspase-independent mechanism. Here, we investigated the mechanisms underlying retinal endothelial cell apoptosis induced by high glucose and oxidative/nitrosative stress conditions. Endothelial cells (TR-iBRB2 rat retinal endothelial cell line) were exposed to high glucose (long-term exposure, 7 days), or to NOC-18 (nitric oxide donor; 250 mu M) or H2O2 (100 mu M) for 24 h. Cell viability was assessed by the MTT assay and cell proliferation by [methyl-H-3]-thymidine incorporation into DNA. Apoptotic cells were detected with Hoechst or Annexin V staining. Active caspases were detected by an apoptosis detection kit. Active caspase-3 and apoptosis-inducing factor (AIF) protein levels were assessed by Western blot or immunohistochemistry. High glucose, NOC-18 and H2O2 increased apoptosis in retinal endothelial cells. High glucose and mannitol decreased cell proliferation, but mannitol did not induce apoptosis. Caspase activation did not increase in high glucose- or NOC-18-treated cells, but it increased in cells exposed to H2O2. However, the protein levels of AIF decreased in mitochondrial fractions and increased in nuclear fractions, in all conditions. These results are the first demonstrating that retinal endothelial cell apoptosis induced by high glucose is independent of caspase activation, and is correlated with AIF translocation to the nucleus. NOC-18 and H2O2 also activate a caspase-independent apoptotic pathway, although H2O2 can also induce caspase-mediated apoptosis. (C) 2008 Elsevier Ltd. All rights reserved.

Heme Oxygenase-1 as Therapeutic Target for Diabetic Foot Ulcers

Dysregulation of endoplasmic reticulum stress response in skin wounds in a streptozotocin-induced diabetes mouse model

Summary: This study investigated the impact of alterations in the unfolded protein response (UPR), autophagy, and mitochondrial function on wound healing (WH) under diabetic conditions. The findings showed that the UPR and autophagy pathways were activated in the skin of control mice and non-wounded skin of diabetic mice. Additionally, high-glucose conditions induced autophagy in keratinocyte and fibroblast cell cultures. However, there were no changes in mitophagy in the skin of diabetic mice or wounded skin.

Summary: Age-related macular degeneration (AMD) is a major cause of visual impairment and blindness in the elderly. The damage to retinal pigment epithelium (RPE) caused by oxidative stress is a key factor in the development of AMD, and this damage is caused by the accumulation of reactive oxygen species (ROS) primarily produced by mitochondria. The protein TRAP1, found in mitochondria, helps maintain mitochondrial integrity and reduces ROS production. This study found that TRAP1 is present in human RPE cells and its levels decrease when exposed to hydrogen peroxide. Silencing TRAP1 leads to increased ROS production and decreased mitochondrial respiratory capacity in RPE cells.

Comparative Analysis of Retinal Organotypic Cultures and In Vivo Axotomized Retinas

Summary: This study compares retinal organotypic cultures (ROCs) with in vivo models to study retinal ganglion cell (RGC) death and glial activation. The results show that there are significant differences in glial activation between ROCs and in vivo models, with earlier and different morphological activation in ROCs. Additionally, microglial cell density in ROCs is consistently lower than in vivo. ROCs can be used for proof-of-concept studies on neuroprotection, but long-term experiments should be performed in vivo.

Sex-specificities in offspring neurodevelopment and behaviour upon maternal glycation: Putative underlying neurometabolic and synaptic changes

Summary: This study aimed to investigate the effects of maternal glycation during lactation on offspring neurodevelopment and behavior, with a focus on sex differences and molecular mechanisms. The researchers found that maternal glycation reduced triglycerides and total antioxidant capacity levels in breast milk. The offspring, both male and female, showed accelerated neurodevelopment milestones during infancy. In adolescence, male offspring exhibited hyperlocomotion, while offspring of both sexes displayed a risk-taking phenotype, accompanied by changes in neural receptor levels. These findings highlight the critical role of the lactation period in metabolic programming and behavioral development.

Self-reported visual function and psychosocial impact of visual loss in EYS-associated retinal degeneration in a Portuguese population

Neonatal testosterone voids sexually differentiated microglia morphology and behavior

Summary: The involvement of immunity in psychiatric disorders, such as anxiety, is characterized by changes in microglia morphology in response to anxiogenic stimuli. Sex differences in microglia morphology and behavior are observed in adolescent rodents, which can be influenced by postnatal testosterone treatment. Administration of testosterone partially eliminates the sex-specific differences in microglia morphology and behavior, approaching towards a male phenotype.

Attention-Deficit/Hyperactivity Disorder Animal Model Presents Retinal Alterations and Methylphenidate Has a Differential Effect in ADHD versus Control Conditions

Summary: Attention-Deficit/Hyperactivity Disorder (ADHD) is a common neurodevelopmental disorder that affects children, and it is associated with ophthalmologic abnormalities. The effects of methylphenidate (MPH) on retinal physiology in ADHD are still unclear. This study used animal models to investigate the structural, functional, and cellular alterations in the retina of ADHD, as well as the impact of MPH. The findings revealed deficits in retinal structure, function, and neurons in the ADHD animal model, and MPH had a beneficial effect on reducing inflammation and blood-retinal barrier dysfunction. However, MPH had opposite effects in the control animals, impairing retinal function and promoting inflammation.

Real-time OXPHOS capacity analysis in wounded skin from diabetic mice: A pilot study

Summary: This study investigated the effects of diabetes on mitochondrial respiration in wounded and non-wounded skin using high-resolution respirometry. The results showed that the oxidative phosphorylation capacity of the wounded skin was decreased at day 3 post-wounding and increased at day 10 post-wounding in diabetic mice. The relative contribution of pyruvate, malate, and glutamate oxidation to the oxidative phosphorylation capacity was higher in wounded skin, while fatty acid oxidation contributed more to the non-wounded skin. This study provides important insights into the impact of diabetes on wound healing.

Regulated cell death pathways in the sodium iodate model: Insights and implications for AMD

Summary: The sodium iodate (NaIO3) model is a valuable tool in studying dry AMD and evaluating the retinal structure. Through extensive research, it has been found that the death of RPE cells caused by NaIO3 injection is closely related to vision loss in patients with dry AMD. Therefore, studying NaIO3 can provide insights into the mechanisms of RPE and photoreceptor cell death and contribute to understanding RPE degeneration in AMD.

A new model of axon degeneration in the mouse optic nerve using repeat intraocular pressure challenge

Summary: We present a new experimental model for inducing retinal ganglion cell (RGC) dysfunction and degeneration in mice. This model allows for the reliable induction of RGC dysfunction and loss, with the resilience of the optic nerve being dependent on the age of the animal. The time-defined nature of the functional and structural changes observed in this model will facilitate the study of glial responses and the evaluation of neuroprotective treatments after intraocular pressure (IOP) injury.
