Summary: Chronic myeloid leukemia (CML) is caused by the fusion tyrosine kinase BCR::ABL1 produced by t(9;22). However, the direct measurement of BCR::ABL1 kinase activity in chronic phase (CP) CML has never been achieved due to degradation in mature leukocytes. In this study, a method was developed to preserve BCR::ABL1 protein in primary CP-CML samples, allowing for the detection of specific kinase activity using a selective substrate. The results showed that BCR::ABL1 kinase activity was barely detectable in CP-CML compared to Ph+ acute lymphoblastic leukemia samples.

Summary: The persistence of leukemic stem cells (LSCs) in chronic myeloid leukemia (CML) presents a challenge in therapy. This study investigates the role of mast cells (MCs) in CML progression and suggests targeting MCs as an additional approach for CML treatment. The BCR::ABL1 fusion gene drives the expansion of bone marrow-derived MCs and enhances their responsiveness to degranulation triggers. In CML patients, splenomegaly is associated with increased BM MC counts and elevated pro-inflammatory cytokines, indicating the importance of MCs in disease progression.

The system also modulates the innate immune response of infected macrophages, enhancing antibacterial capacity.

Summary: This study aimed to explore the influences of SAP2 and CAP1 on itraconazole (ITR) resistance of Candida albicans at different states. The results showed that the biofilm formation capacity of resistant C. albicans strains, including the CAP1 Delta/Delta strain, was stronger. The expression of SAP2 and CAP1 were both significantly upregulated in resistant strains compared to sensitive strains, and CAP1 knockdown increased SAP2 expression. These findings suggest that loss of CAP1 can increase SAP2 level and influence biofilm formation, leading to increased ITR resistance in C. albicans.

Summary: The intracellular survival of pathogenic bacteria relies on various survival strategies and virulence factors. However, treatment of these infections often fails due to challenges such as poor antibiotic penetration and retention in host cells. Drug delivery systems (DDSs) have shown promise in overcoming these limitations and improving the effectiveness of antibiotic therapy. This review provides an overview of the classification and mechanisms of intracellular bacterial persistence, describes the design strategies used for DDSs to eliminate intracellular bacteria, and highlights the approaches for internalization, bacterial targeting, and immune enhancement. It offers guidance for developing functional DDSs to effectively eradicate intracellular bacteria.

Summary: This study assessed the influence of di-n-butyl phthalate (DBP) on the multi-functionality of composting processes. The results showed that DBP stress negatively affected multi-functionality, resulting in a 6.5% reduction. Additionally, DBP stress led to decreased microbial biomass, altered microbial composition, and decreased network complexity. Keystone taxa, including Streptomyces, Thermoactinomyces, Mycothermus, and Lutispora, were found to be significantly affected by DBP stress and showed a correlation with multifunctionality. This study contributes to a better understanding of the negative implications of phthalates during composting processes and provides insights for the development of new treatment strategies for agricultural waste.
