miR-29b suppresses CML cell proliferation and induces apoptosis via regulation of BCR/ABL1 protein

MicroRNAs (miRNAs) are small RNAs that regulate gene expression posttranscriptionally and are critical for many cellular pathways. Recent evidence has shown that aberrant miRNA expression profiles and unique miRNA signaling pathways are present in many cancers. Here, we demonstrate that miR-29b is markedly lower expressed in CML patient samples. Bioinformatics analysis reveals a conserved target site for miR-29b in the 3'-untranslated region (UTR) of ABL1. miR-29b significantly suppresses the activity of a luciferase reporter containing ABL1-3'UTR and this activity is not observed in cells transfected with mutated ABL1-3'UTR. Enforced expression of miR-29b in K562 cells inhibits cell growth and colony formation ability thereby inducing apoptosis through cleavage of procaspase 3 and PARP. Furthermore, K562 cells transfected with a siRNA targeting ABL1 show similar growth and apoptosis phenotypes as cells overexpression of miR-29b. Collectively, our results suggest that miR-29b may function as a tumor suppressor by targeting ABL1 and BCR/ABL1. (c) 2013 Elsevier Inc. All rights reserved.

Effects of combined soil amendments on Cd accumulation, translocation and food safety in rice: a field study in southern China

Summary: The study investigated the reduction effects of combined soil amendments on Cd content and Cd/Zn ratio in rice grains in soils with different Cd contamination levels in three regions of Hunan province, China. The combined amendments, especially lime combined with organic fertilizer, proved to be effective in reducing Cd content in rice grains and Cd/Zn ratio, as well as decreasing soil available Cd and Zn concentration.

Cascade-Targeting Poly(amino acid) Nanoparticles Eliminate Intracellular Bacteria via On-Site Antibiotic Delivery

Summary: A cascade-targeting drug delivery system has been developed to sequentially target macrophages and intracellular bacteria, allowing on-site drug delivery. The system encapsulates rifampicin into mannose-decorated nanoparticles, which are designed to specifically target macrophages and then release the drug in acidic environments to effectively eliminate intracellular bacteria. The system also modulates the innate immune response of infected macrophages, enhancing antibacterial capacity.

Source apportionment of environmentally persistent free radicals (EPFRs) and heavy metals in size fractions of urban arterial road dust

Summary: The study investigated the pollution characteristics and sources of heavy metals and EPFRs in mixed particle-size road dust samples from urban arterial roads in three cities. Results showed that a significant portion of road dust consisted of <75 μm particles, with high levels of EPFRs and heavy metals in smaller size fractions. Traffic emissions were identified as the main source for Cr, Pb, Zn, and EPFRs, while industrial activities were the primary source of Cd and Cu in fine particle fractions.

Correlation between SAP2 and CAP1 in clinical strains of Candida albicans at planktonic and biofilm states

Summary: This study aimed to explore the influences of SAP2 and CAP1 on itraconazole (ITR) resistance of Candida albicans at different states. The results showed that the biofilm formation capacity of resistant C. albicans strains, including the CAP1 Delta/Delta strain, was stronger. The expression of SAP2 and CAP1 were both significantly upregulated in resistant strains compared to sensitive strains, and CAP1 knockdown increased SAP2 expression. These findings suggest that loss of CAP1 can increase SAP2 level and influence biofilm formation, leading to increased ITR resistance in C. albicans.

Targeted Drug Delivery Systems for Eliminating Intracellular Bacteria

Summary: The intracellular survival of pathogenic bacteria relies on various survival strategies and virulence factors. However, treatment of these infections often fails due to challenges such as poor antibiotic penetration and retention in host cells. Drug delivery systems (DDSs) have shown promise in overcoming these limitations and improving the effectiveness of antibiotic therapy. This review provides an overview of the classification and mechanisms of intracellular bacterial persistence, describes the design strategies used for DDSs to eliminate intracellular bacteria, and highlights the approaches for internalization, bacterial targeting, and immune enhancement. It offers guidance for developing functional DDSs to effectively eradicate intracellular bacteria.

Di-n-butyl phthalate stress hampers compost multifunctionality by reducing microbial biomass, diversity and network complexity

Summary: This study assessed the influence of di-n-butyl phthalate (DBP) on the multi-functionality of composting processes. The results showed that DBP stress negatively affected multi-functionality, resulting in a 6.5% reduction. Additionally, DBP stress led to decreased microbial biomass, altered microbial composition, and decreased network complexity. Keystone taxa, including Streptomyces, Thermoactinomyces, Mycothermus, and Lutispora, were found to be significantly affected by DBP stress and showed a correlation with multifunctionality. This study contributes to a better understanding of the negative implications of phthalates during composting processes and provides insights for the development of new treatment strategies for agricultural waste.

Low serum dehydroepiandrosterone levels are associated with diabetic retinopathy in patients with type 2 diabetes mellitus

Summary: This cross-sectional study in China found a significant association between low serum dehydroepiandrosterone levels and diabetic retinopathy in patients with type 2 diabetes mellitus. The study also showed a linear decrease in the odds of diabetic retinopathy with increasing dehydroepiandrosterone concentration. Subgroup analyses indicated that dehydroepiandrosterone level consistently affected diabetic retinopathy.

Therefore, this dual-targeted DDS-assisted intracellular synergistic antibacterial strategy is an effective therapeutic approach against intracellular bacteria.
