Diabetic Complications in Obese Type 2 Diabetic Rat Models

We overviewed the pathophysiological features of diabetes and its complications in obese type 2 diabetic rat models: Otsuka Long-Evans Tokushima fatty (OLETF) rat, Wistar fatty rat, Zucker diabetic fatty (ZDF) rat and Spontaneously diabetic Toni (SDT) fatty rat. Pancreatic changes with progression of diabetes were classified into early changes, such as islet hypertrophy and degranulation of 13 cells, and degenerative changes, such as islet atrophy and fibrosis of islet with infiltration of inflammatory cells. Renal lesions in tubuli and glomeruli were observed, and nodular lesions in glomeruli were notable changes in OLETF and SDT fatty rats. Among retinal changes, folding and thickening were interesting findings in SDT fatty rats. A decrease of motor nerve conduction velocity with progression of diabetes was presented in obese diabetic rats. Other diabetic complications, osteoporosis and sexual dysfunction, were also observed. Observation of bone metabolic abnormalities, including decrease of osteogenesis and bone mineral density, and sexual dysfunction, including hypotestosteronemia and erectile dysfunction, in obese type 2 diabetic rats have been reported.

Pathophysiological features in the brains of female Spontaneously Diabetic Torii (SDT) fatty rats

Summary: Diabetes mellitus (DM) and obesity are associated with neurodegenerative diseases and psychiatric disorders. This study investigated pathophysiological changes in the brain of female SDT fatty rats with diabetes and obesity. The results showed decreased cell numbers and increased expression of an inflammation-related gene in the brains of SDT fatty rats.

Association analysis between novel variants in LEPR gene and litter size in Mongolia and ujimqin sheep breeds

Summary: In this study, novel variations in the LEPR gene were identified in Mongolia sheep, and two mutations were significantly associated with litter sizes in Mongolia and Ujimqin ewes. These mutations are predicted to alter the mRNA secondary structure of LEPR. The findings provide potential genetic markers for increasing litter size in sheep.

Immune Disorders and Sex Differences in Spontaneously Diabetic Torii Rats, Type 2 Diabetic Model

Summary: Type 2 diabetes is a non-autoimmune metabolic disorder, but there are increasing reports of immune abnormalities in T2D patients. This study suggests that SDT rats may be a useful T2D model involving immune abnormalities, and immune abnormalities in male SDT rats may contribute to the development of diabetes.

The role of BBS2 in regulating adipogenesis and the association of its sequence variants with meat quality in Qinchuan cattle

Summary: This study explores the role of the BBS2 gene in fat deposition of beef cattle and identifies SNPs associated with meat quality traits. It finds that the expression of BBS2 is negatively correlated with marbling ratios in beef cattle. Knockdown of BBS2 promotes adipogenesis and lipid accumulation in bovine preadipocytes. Several SNPs in the exons of BBS2 are significantly associated with meat quality in Chinese Qinchuan cattle. These findings have implications for improving meat quality and breeding in beef cattle.

Establishment of a new nonalcoholic steatohepatitis model; Ovariectomy exacerbates nonalcoholic steatohepatitis-like pathology in diabetic rats

Summary: An increasing number of patients worldwide are being diagnosed with nonalcoholic fatty liver disease and nonalcoholic steatohepatitis (NAFLD/NASH) due to the growing prevalence of obesity and metabolic disorders. The incidence of NAFLD is higher in postmenopausal women than in premenopausal women, and the decline in female hormone levels may contribute to the deterioration of metabolism. In this study, a new animal model for NAFLD was investigated using Spontaneously Diabetic Torii (SDT) fatty rats. The results showed that the OVX-SDT fatty rats exhibited NASH with more severe hepatic fibrosis compared to the untreated animals, suggesting that estrogen reduction induced by ovariectomy may enhance lipid synthesis in the liver.

Age-dependent changes of hindgut microbiota succession and metabolic function of Mongolian cattle in the semi-arid rangelands

Summary: This study investigated the bacterial community and associated metabolome in the cecum and colon of Mongolian cattle at different ages after weaning. The results showed the specific development patterns of hindgut microorganisms and the dynamic changes of metabolites. The findings highlighted the importance of microbial and metabolite interactions in enhancing the abiotic stress resistance of Mongolian cattle in a harsh environment.

Effects of the Expressions and Variants of the CAST Gene on the Fatty Acid Composition of the Longissimus Thoracis Muscle of Grazing Sonid Sheep

Summary: This study aimed to evaluate the relationship between the expression levels of the CAST gene and the fatty acid (FA) composition in the longissimus thoracis (LL) muscle and to identify novel variants of CAST and perform association analysis with the FA composition in grazing Sonid sheep. The correlation results showed that high expression levels of CAST are correlated with better FA compositions and classes in LL. Four mutations were identified in the CAST gene of Sonid sheep. The association studies showed that these mutations are associated with specific FA compositions and classes in the LL of Sonid sheep, indicating their potential as genetic selection markers for meat quality.

Unilateral Nephrectomized SHR/NDmcr-cp Rat Shows a Progressive Decline of Glomerular Filtration With Tubular Interstitial Lesions

Summary: In patients with diabetic kidney disease (DKD), there is a need for animal models to evaluate renal function decline and assess the efficacy of therapeutic agents. This study developed a DKD animal model using spontaneously hypertensive rat (SHR)/NDmcr-cp (cp/cp) rats with characteristics of obese type 2 diabetes. Unilateral nephrectomy (UNx) in these rats resulted in a decline in creatinine clearance rate (Ccr), glomerular sclerosis, tubular lesions, and tubulointerstitial fibrosis, accompanied by renal anemia. Treatment with losartan-mixed diet suppressed Ccr decline, improved renal anemia and histopathological changes. These findings suggest that UNx-SHR/cp rats can be used as a DKD model for evaluating therapeutic agents.

Effects of novel variants in BMP15 gene on litter size in Mongolia and Ujimqin sheep breeds

Summary: Reproductive traits are important in the sheep industry, and the BMP15 gene plays a major role. This study identified 12 novel variants in the BMP15 gene of Mongolia sheep, and found significant associations between certain variants and litter size in Mongolia ewes and Ujimqin ewes. One of the mutations is predicted to affect the tertiary structure of BMP15. These findings may contribute to improving litter size in sheep.

Summary: This study evaluated the relevance of an animal model to human nonalcoholic fatty liver disease (NAFLD) by gene expression analysis. The results showed that a choline-deficient L-amino acid-defined (CDAA) diet-induced NAFLD animal model successfully recaptured key aspects of human pathophysiology, particularly in terms of immune cell functions.
