Summary: The cross-industry survey conducted in 2018 revealed that a staged approach for phototoxicity assessment has been widely accepted globally, with the OECD-based 3T3 NRU Phototoxicity Test being the most frequently used in vitro method. The survey also showed that in vitro-positive results allowed further development without additional data, and that in vivo phototoxicity studies lack formal guidelines and standardization across companies. Finally, a margin-of-safety approach based on Cmax at NOAEL has successfully supported clinical development.

Summary: This study describes the varying degrees of phototoxicity in fluorescent imaging agents that are not intended to be phototoxic. Several fluorescent agents were evaluated for their phototoxicity in vitro and compared to known phototoxic agents. The results indicate that IRdye800 and ICG are relatively safe, while precautions need to be taken when using proflavine and methylene blue as fluorescent imaging agents due to their potential phototoxicity.

Summary: The phototoxic potential of commonly used tattoo ink colors was examined in this study. The results showed that the tested tattoo inks did not exhibit cytotoxic activity or phototoxic effects. However, further testing methods are needed to comprehensively evaluate the phototoxic complications of different tattoo inks.

Summary: The phototoxic potential of furocoumarins has been established, but bergamottin, which contains a bulky, hydrophobic side chain, has significantly lower or even no phototoxicity potential. The OECD Test Guideline 432 3T3/Neutral Red Uptake (NRU) in vitro phototoxicity test is a reliable method for identifying compounds with no phototoxicological potential. In this study, bergamottin showed no phototoxic potential, while established phototoxic furocoumarins 5-MOP, 8-MOP, and psoralen exhibited phototoxicity. Bergamottin had negative results at higher molar-adjusted concentrations compared to 5-MOP, 8-MOP, and psoralen.

Summary: The increased use of agrochemicals raises concerns about environmental, animal, and mainly human toxicology. New Approach Methodologies (NAMs) including in vitro tests and in silico protocols are encouraged for toxicological risk assessment. In this study, in vitro and in silico techniques were integrated to assess the mutagenic and phototoxic potential of agrochemicals. The results showed that the tested agrochemicals did not exhibit mutagenic potential in the two proposed approaches.

Summary: Melanoma is a dangerous cancer with aggressive properties and low effectiveness of current therapies. Tetracyclines have shown anticancer activity, and this study analyzes their phototoxic effect on melanoma cells. The results demonstrate that tetracycline-induced phototoxicity significantly reduces the number of live cells by cell cycle arrest and decreased cell viability.

Summary: This study evaluated the influence of Meloxicam and UVAR on skin cell homeostasis. The results showed that co-treatment with Meloxicam and UVAR inhibited skin cell proliferation and caused morphological changes. Additionally, Meloxicam and UVAR disrupted redox homeostasis by lowering the intracellular level of reduced thiols. The study revealed that the phototoxic activity of Meloxicam is associated with oxidative stress induction and disruptions in cell homeostasis.
