Preventive effect of aminoguanidine compared to vitamin E and C on cisplatin-induced nephrotoxicity in rats

In this study. the antioxidant effect of aminoguanidine on nephrotoxicity of a single dose of cisplatin is investigated and compared with the effects of well-known antioxidants vitamin C and E combination. Tubular damage and perivascular inflammation were observed in kidney samples of the cisplatin-administered groups. Aminoguanidine and vitamin C-E combination are found to be capable of preventing these effects of cisplatin. Liver tissues Of all groups were intact. Cisplatin-induced oxidative stress was evidenced by significant decrease in glutathione and significant increase in malondialdehyde levels in kidney samples. Antioxidants with cisplatin decreased malondialdehyde levels. Antioxidants with cisplatin prevented the decrease in liver glutathione levels. The nephrotoxicity was confirmed biochemically, by significant elevation of serum urea and creatinine levels. Both vitamin C-E combination and aminoguanidine prevented the increase in serum urea levels according to the cisplatin group. (C) 2008 Elsevier GmbH. All rights reserved.

Bioactivation of clozapine by mitochondria of the murine heart: Possible cause of cardiotoxicity

Summary: The study suggests that clozapine-induced cardiotoxicity may be caused by the activation of cardiac mitochondria. Mitochondria in cardiac tissue contain CYP enzymes that can catalyze the formation of reactive metabolites from clozapine. Clozapine significantly reduces mitochondrial oxygen consumption rate, providing an explanation for its adverse effects on the heart.

In vitro evaluation of estrogenic, antiestrogenic and antitumor effects of amentoflavone

Summary: Apigenin, a flavonoid, is known to act as an estrogen receptor agonist and inhibit aromatase enzyme. Amentoflavone, a biflavonoid, has been evaluated for its endocrine modulatory effects and antitumor activity in breast cancer cells, showing potential selective cytotoxicity but not a viable alternative to current drugs. Further studies are required to investigate its in vivo effects.

Summary: Oxidative stress disrupts the balance between prooxidants and antioxidants, leading to potential biomolecular damage and involvement in various diseases. NRF2, a key regulator of oxidative stress, has potential benefits in the prevention of breast cancer but overactivation may negatively impact cancer therapy. Known oxidative stress modulators like melatonin and polyphenols are suggested to play a role in cancer prevention and treatment through NRF2 activation.

Mitochondrial versus microsomal bioactivation of paracetamol by human liver and kidney tissues

Summary: Mitochondria are the major target of paracetamol-induced hepatotoxicity. This study found that mitochondrial CYPs can catalyze the formation of toxic metabolite NAPQI from paracetamol, leading to the transmission of death signals. However, in the kidneys, there is no correlation between the formation of NAPQI and kidney CYP2E1 activity, suggesting a different toxicity mechanism compared to the liver.

Role of Oxidative Stress and Reactive Metabolites in Cytotoxicity & Mitotoxicity of Clozapine, Diclofenac and Nifedipine in CHO-K1 Cells in vitro

Summary: This study investigated the cytotoxic mechanisms of clozapine, diclofenac, and nifedipine using CHO-K1 cells. It was found that the cytotoxicity of nifedipine and clozapine could be decreased by soft and hard nucleophilic agents, while diclofenac's cytotoxicity was paradoxically increased. Furthermore, oxidative stress and membrane damage were observed in all three drugs, with diclofenac and nifedipine causing mitochondrial membrane damage. These findings suggest that these drugs may exert cytotoxic effects through different mechanisms.
