Ethanolic Extract of Propolis Augments TRAIL-Induced Apoptotic Death in Prostate Cancer Cells

Prostate cancer is a commonly diagnosed cancer in men. The ethanolic extract of propolis (EEP) and its phenolic compounds possess immunomodulatory, chemopreventive and antitumor effects. Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL/APO2L) is a naturally occurring anticancer agent that preferentially induces apoptosis in cancer cells and is not toxic to normal cells. We examined the cytotoxic and apoptotic effects of EEP and phenolic compounds isolated from propolis in combination with TRAIL on two prostate cancer cell lines, hormone-sensitivity LNCaP and hormone-refractory DU145. The cytotoxicity was evaluated by MTT and LDH assays. The apoptosis was determined using flow cytometry with annexin V-FITC/propidium iodide. The prostate cancer cell lines were proved to be resistant to TRAIL-induced apoptosis. Our study demonstrated that EEP and its components significantly sensitize to TRAIL-induced death in prostate cancer cells. The percentage of the apoptotic cells after cotreatment with 50 mu gmL(-1) EEP and 100 ngmL(-1) TRAIL increased to 74.9 +/- 0.7% for LNCaP and 57.4 +/- 0.7% for DU145 cells. The strongest cytotoxic effect on LNCaP cells was exhibited by apigenin, kaempferid, galangin and caffeic acid phenylethyl ester (CAPE) in combination with TRAIL (53.51 +/- 0.68-66.06 +/- 0.62% death cells). In this work, we showed that EEP markedly augmented TRAIL-mediated apoptosis in prostate cancer cells and suggested the significant role of propolis in chemoprevention of prostate cancer.

The comparative analysis of selected interleukins and proinflammatory factors in CSF among de novo diagnosed patients with RRMS

Summary: This study aimed to assess the levels of selected interleukins and proinflammatory factors in cerebrospinal fluid (CSF) among patients diagnosed with relapsing-remitting multiple sclerosis (RRMS). The results showed that IL-5 and IL-13 levels increased with the age at diagnosis of MS, while the concentration of IL-10 was lower in patients over the age of 35. The levels of IFN-gamma, TNF-alpha, IL-5, IL-10 and IL-15 increased with the longer time from the first symptoms to diagnosis. IL-2 levels were significantly higher among patients without gadolinium-enhanced (Gd(+)) MRI lesions.

The study provides valuable insights into the role of HHLA2 as a stimulatory and inhibitory immune checkpoint in CRC and suggests the potential therapeutic value of the HHLA2-KIR3DL3/TMIGD2 pathway.

B7H3 Role in Reshaping Immunosuppressive Landscape in MSI and MSS Colorectal Cancer Tumours

Summary: Colorectal cancer (CRC) is a common malignant tumor with a high mortality rate. This study found that B7H3 is upregulated in CRC tumors and correlated with M2-macrophage polarization and tumor growth-promoting cytokines. Additionally, high B7H3 expression is associated with decreased survival in CRC patients. These findings provide new insights into the role of B7H3 in CRC immunity.

Summary: Successful implantation in augmented areas requires adequate bone density and quality, as well as thorough planning. This study evaluated the healing process after implant surgery (dental implants) using dynamic surgical navigation following bone augmentation. The results showed faster healing in patients treated with the flapless technique and Navident dynamic navigation, along with lower levels of pro-inflammatory cytokines.

Summary: Inflammation plays a crucial role in immune defense, but chronic inflammation may lead to neurodegenerative diseases, atherosclerosis, inflammatory bowel diseases, or cancer. Flavanones found in citrus fruits possess anti-oxidative and anti-inflammatory properties. This study demonstrates that methyl derivatives of flavanone can inhibit inflammation and modulate the production of inflammatory cytokines.

The findings suggest that certain molecules may be promising biomarkers for MS, but further research is needed to understand their precise functions.
