Summary: This study investigates the role of signal transducer and activator of transcription 4 (STAT4) in neutrophils during advanced atherosclerosis. STAT4 deficiency reduces plaque burden and improves plaque stability in mice. Additionally, STAT4 deficiency decreases neutrophil activation and attenuates neutrophil trafficking to atherosclerotic plaques.

Summary: Atherosclerosis is a chronic inflammatory disease that involves the recruitment and differentiation of monocytes into macrophages in response to inflammatory insults. Macrophages play critical roles in tissue damage, wound healing, and regression of atherosclerotic lesions, and their phenotypes are influenced by various stimuli. Recent advances in single-cell sequencing have revealed the heterogeneity of macrophages in lesions, shedding light on potential macrophage-based therapies for atherosclerosis.

Summary: IL-6 trans-signaling regulates the expression of laminin alpha chains in endothelial cells and inhibits the migration of granulocytic cells across endothelial cells. The expression of laminin alpha chains is altered in human atherosclerotic plaques and is related to the abundance of leukocyte subpopulations.

Summary: This study shows that deficiency of bilirubin is associated with increased risk of atherosclerosis and cardiovascular disease. In the experiments, mice deficient in bilirubin showed higher burden of atherosclerotic plaques, increased systemic oxidative stress, endothelial dysfunction, and hyperlipidemia. Furthermore, the study found that bilirubin deficiency selectively enhanced neutrophil-mediated inflammation and destabilization of unstable plaques.

Summary: This study demonstrates that circulating IL-6 levels predict carotid plaque severity, vulnerability, and progression. A cutoff of 2.0 pg/mL for IL-6 levels could facilitate the selection of individuals that would benefit from anti-IL-6 drugs for stroke prevention.

Summary: According to the WHO, the entire global population is exposed to air pollution levels higher than recommended for health preservation. Air pollution, a complex mixture of particles and gases, poses a major global threat to public health. Particulate matter PM2.5 has been identified as a causal factor for cardiovascular diseases such as hypertension, coronary artery disease, stroke, heart failure, arrhythmias, and cardiovascular mortality. This review explores the proatherogenic effects of PM2.5, including endothelial dysfunction, chronic inflammation, reactive oxygen species, mitochondrial dysfunction, and arterial plaque instability.

Summary: The research suggests that serum IRGM levels may be related to plaque rupture in STEMI patients, and deficiency of IRGM/Irgm1 can increase plaque stability by suppressing macrophage apoptosis through inhibiting ROS generation and MAPK signaling transduction. Targeting IRGM may represent a new treatment strategy for the prevention and treatment of acute cardiovascular deaths caused by plaque rupture.

Summary: Despite efforts to develop new therapies, atherosclerotic cardiovascular diseases (ASCVD) remain a leading cause of death worldwide. However, current approaches to prognosis, diagnosis, and classification of acute coronary syndrome do not consider the diversity of plaque phenotypes.

Summary: Atherosclerosis and its complications are major causes of cardiovascular morbidity and mortality globally. The Yes-associated protein (YAP) has been shown to play a crucial role in the pathogenesis and development of atherosclerosis, regulating processes such as vascular dysfunction, monocyte infiltration, and smooth muscle cell migration. Further research is needed to explore the role of YAP in atherosclerotic complications like vascular calcification and intraplaque hemorrhage.

Summary: Arteriovenous fistulas (AVFs) are commonly used for hemodialysis, but their failure rate is high. Abnormal proliferation and migration of vascular smooth muscle cells along with increased mitochondrial fission may contribute to AVF stenosis. Inhibiting mitochondrial fission could potentially improve AVF patency.

Summary: Chronic thromboembolic pulmonary hypertension (CTEPH) is characterized by thrombofibrotic obstruction of the proximal pulmonary arteries, leading to vascular remodeling of the distal pulmonary artery. The study identified molecular mechanisms, with TNF-alpha driving CTEPH pathogenesis by promoting VSMC proliferation and migration through increased FOS expression. This information may guide the development of new therapeutic targets for CTEPH.
