Impact of atomization technique on the stability and transport efficiency of nebulized liposomes harboring different surface characteristics

The objective of this study was to evaluate the impact of nebulization on liposomes with specific surface characteristics by applying three commercially available inhaler systems (air-jet, ultrasonic and vibrating-mesh). Conventional liposome formulations composed of phosphatidylcholine and cholesterol were compared to sterically stabilized PEGylated liposomes and cationic polymer coated liposomes. Liposomes of similar size (between 140 and 165 nm in diameter with polydispersity indices <0.1) were prepared by dry lipid film rehydration followed by size extrusion. Their stability upon nebulization was determined in terms of size, polydispersity index and leakage using a fluorescence quenching system. The transport efficiencies of the nebulizer devices and the influences of both salt and liposomes on the droplet size distribution of the aerosol were investigated. While the droplet size of the aerosol decreased with increasing salt concentration the liposomes had no influence on the droplet size distribution. The output of the nebulizers in terms of liposomal transport efficiencies differed significantly among the nebulizer principles (20-100%, p < 0.05), with the vibrating-mesh nebulizers being the most effective. The integrity of the conventional liposomes was almost unaffected by the atomization process, while polymer coated and especially positively charged liposomes showed enhanced leakage. The release rates for the hydrophilic model drug system were highest for the vibrating-mesh nebulizers regardless of the surface characteristics of the liposomes (increasing from 10% to 20% and 50% for the conventional, PEGylated and positively charged formulations, respectively). In view of surface modified liposomes our data suggest that drug delivery via nebulization necessitates the finding of a compromise between nebulizer efficiency, formulation stability and drug release profile to accomplish the development of tailored formulations suitable for advanced inhalation therapy. Divergent Roles of Ephrin-B2/EphB4 Guidance System in Pulmonary Hypertension

Summary: Through this study, it was found that ephrin-B2 and EphB4 expression is preserved in smooth muscle and endothelial cells of remodeled pulmonary arteries. Lack of ephrin-B2 gene attenuates pulmonary vascular remodeling and right ventricular hypertrophy in chronic hypoxia-induced pulmonary hypertension, while inhibition of EphB4 signaling reduces pulmonary vascular remodeling without affecting right ventricular systolic pressure. In conclusion, ephrin-B2 and EphB4 play crucial roles in pulmonary vascular remodeling.

Identification of extracellular vesicle microRNA signatures specifically linked to inflammatory and metabolic mechanisms in obesity-associated low type-2 asthma

Summary: This study aimed to use EV miRNA signatures to uncover the molecular mechanisms of obesity-associated low type-2 asthma. EV miRNA profiles were able to clearly distinguish between obese low type-2 asthma, non-obese low type-2 asthma, and healthy control groups. Pathway analysis revealed inflammatory cytokines and metabolic factors signaling pathways to be specifically associated with obesity-associated low type-2 asthma. Additionally, miR-17-92 and miR-106a-363 clusters were significantly enriched in obesity-associated low type-2 asthma and exhibited correlations with key laboratory and lung function parameters.

Lipid Nanoparticles as a Shuttle for Anti-Adipogenic miRNAs to Human Adipocytes

Summary: Obesity and type 2 diabetes are significant health burdens without effective treatment options. This study explores the use of PEG-LNPs to deliver anti-adipogenic miRNAs (miR26a and miR27a) in order to regulate gene expressions and balance adipose tissue metabolic functions. The findings demonstrate that upregulating miR27a inhibits adipogenesis in preadipocytes, while transfecting miR26a induces white adipocyte browning. Selective delivery of miRNAs to adipocytes through PEG-LNPs may offer new possibilities for treating obesity and related metabolic diseases.

Summary: The role of pulmonary vasculature in the development and progression of acute and chronic lung diseases, such as ARDS, PF, and COPD, has been frequently overlooked. However, there is increasing evidence that the vascular endothelium plays an active role in these diseases. This review outlines the current knowledge and future research directions regarding the pulmonary vasculature and hopes to facilitate the development of mechanism-based therapies.
