Immediate release pellets with lipid binders obtained by solvent-free cold extrusion

Lipid-based drug delivery systems have spread in their use in pharmaceutical drug development. This work focuses on the use of lipid binders as alternative non-toxic extrusion aid for pellet formulations. The preparation of immediate release pellets with solid lipid binders through a solvent-free cold extrusion/spheronisation process was investigated in this study. Various binary, ternary and quaternary mixtures of powdered lipids and the model drug sodium benzoate were investigated and compared to well-known wet extrusion binders like microcrystal line cellulose and K-carrageenan. The cold lipid extrusion process offers multiple advantages as it is suitable for thermal sensitive as well as for hygroscopic drugs, furthermore no drying process to evaporate the solvent is needed and the process is feasible for different extruder types. Some of the developed pellets showed favourable properties like spherical shape, narrow size distribution, a high drug load of 80% sodium benzoate and a drug release of more than 90% within 40 min. The stability of drug release, which can be problematic when using lipid excipients, was sufficient for some mixtures, as storage under elevated temperatures changed the release profiles only slightly and no formulation released less than 80% within the first 60 min. A formulation with a mixture of hard fat, glycerol distearate and glycerol trimyristate showed the best results, as pellets with a low aspect ratio, narrow size distribution and complete drug release were obtained. Using appropriate mixtures of acylglycerides it becomes possible to produce pharmaceutical pellets with immediate release characteristics by cold extrusion and subsequent spheronisation. Precipitation from amorphous solid dispersions in biorelevant dissolution testing: The polymorphism of regorafenib

Application and validation of a coaxial liquid core waveguide fluorescence detector for the permeation analysis of desmopressin acetate

Summary: The development of peptide drugs and alternative routes of administration has become increasingly important to the pharmaceutical industry in recent years. A new HPLC-coupled coaxial liquid-core waveguide fluorescence detector has been developed and evaluated for sensitive detection of large molecules and peptides. The validation results showed increased sensitivity, with a limit of detection of 4.7 ng/mL and a lower limit of quantification of 9.5 ng/mL, demonstrating successful observation of desmopressin permeation in clinically relevant dosages and time periods.

Transfer and scale-up of the manufacturing of orodispersible mini-tablets from a compaction simulator to an industrial rotary tablet press

Summary: The study demonstrated the feasibility of using isomalt and Ludiflash (R) for the development of ODMTs, in accordance with the guidelines of the FDA and EMA. ODMTs produced on a compaction simulator and a rotary tablet press showed similar characteristics and texture, but during scale-up process, the disintegration time significantly increased.

Precise Dosing of Pramipexole for Low-Dosed Filament Production by Hot Melt Extrusion Applying Various Feeding Methods

Summary: The aim of this research was to produce low-dosed filaments via hot-melt extrusion (HME) with a model drug for Parkinson's disease treatment. It was found that deviation from target drug concentration occurred due to degradation products in certain formulations. Material temperature and the formation of anhydrous drug in the extruded filaments should be considered for the calculation of recovered API. The study showed that equilibrium condition for drug content was reached relatively late in the process, and the calculated RSD by Stange-Poole equation can be used to evaluate the deviations of drug content in equilibrium conditions.

This requires both innovative drug products and new analytical methods for drug development and quality control. The liquid-core waveguide UV absorbance flow cell detector coupled to an existing HPLC-UV system allows for simultaneous determination of different active ingredients and wide concentration ranges, increasing sensitivity and reducing analysis time.
