Enduring increases in anxiety-like behavior and rapid nucleus accumbens dopamine signaling in socially isolated rats

Social isolation (SI) rearing, a model of early life stress, results in profound behavioral alterations, including increased anxiety-like behavior, impaired sensorimotor gating and increased self-administration of addictive substances. These changes are accompanied by alterations in mesolimbic dopamine function, such as increased dopamine and metabolite tissue content, increased dopamine responses to cues and psychostimulants, and increased dopamine neuron burst firing. Using voltammetric techniques, we examined the effects of SI rearing on dopamine transporter activity, vesicular release and dopamine D2-type autoreceptor activity in the nucleus accumbens core. LongEvans rats were housed in group (GH; 4/cage) or SI (1/cage) conditions from weaning into early adulthood [postnatal day (PD) 2877]. After this initial housing period, rats were assessed on the elevated plus-maze for an anxiety-like phenotype, and then slice voltammetry experiments were performed. To study the enduring effects of SI rearing on anxiety-like behavior and dopamine terminal function, another cohort of similarly reared rats was isolated for an additional 4months (until PD 174) and then tested. Our findings demonstrate that SI rearing results in lasting increases in anxiety-like behavior, dopamine release and dopamine transporter activity, but not D2 activity. Interestingly, GH-reared rats that were isolated as adults did not develop the anxiety-like behavior or dopamine changes seen in SI-reared rats. Together, our data suggest that early life stress results in an anxiety-like phenotype, with lasting increases in dopamine terminal function.

Blocking voltage-gated K+ channels with 4-aminopyridine had the greatest effects in ventral regions of female mice, indicating regional differences in Kv channel expression. The dopamine transporter blocker cocaine enhanced detection across subregions in both sexes, with females showing greater overall increased release than males.

Enhanced heroin self-administration and distinct dopamine adaptations in female rats

Summary: Evidence suggests that females are more vulnerable to the harmful effects of drugs like opioids, with increased heroin-related deaths seen in females. While males and females showed similar acquisition rates for heroin self-administration (SA), females exhibited increased responding and intake, seeking behavior, and escalation. The study found no differences in mu-opioid receptor (MOR) expression levels between males and females in the brain, but observed differential dopamine terminal adaptations in females that may contribute to their increased heroin-seeking behavior.

Kappa Opioid Receptors Reduce Serotonin Uptake and Escitalopram Efficacy in the Mouse Substantia Nigra Pars Reticulata

Summary: The serotonin and kappa opioid receptor (KOR) systems play important roles in disorders of negative affect. In this study, the researchers investigated the effects of KOR activation and serotonin reuptake inhibition on serotonin transmission in the substantia nigra pars reticulata (SNr). They found that KOR activation reduced serotonin release and uptake, while serotonin reuptake inhibition increased serotonin release and slowed reuptake. The researchers also demonstrated that short-term activation of KOR altered serotonin responses to reuptake inhibition. These findings highlight the complex interactions between KORs and serotonin in regulating negative affect, and suggest that the KOR system may be a promising target for treating disorders such as anxiety and depression alongside traditional antidepressant treatments.

Kappa opioid receptor agonist U50,488 inhibits dopamine more in caudal than rostral nucleus accumbens core

Summary: The nucleus accumbens (NAc) core plays a role in regulating stress and shaping reward seeking behaviours. The activation of kappa opioid receptor (KOR) inhibits dopamine release and transmission, and this impact is regionally specific in the NAc shell. The inhibition of dopamine release by KOR is greater in the caudal compared to the rostral subregion of NAc core, suggesting a differential regulation along the rostro-caudal axis.
