Diet-dependent modulation of hippocampal expression of endocannabinoid signaling-related proteins in cannabinoid antagonist-treated obese rats

Diet-induced obesity produces changes in endocannabinoid signaling (ECS), influencing the regulation of energy homeostasis. Recently, we demonstrated that, in high-fat-diet-fed rats, blockade of CB1 receptor by AM251 not only reduced body weight but also increased adult neurogenesis in the hippocampus, suggesting an influence of diet on hippocampal cannabinoid function. To further explore the role of hippocampal ECS in high-fat-diet-induced obesity, we investigated whether the immunohistochemical expression of the enzymes that produce (diacylglycerol lipase alpha and N-acyl phosphatidylethanolamine phospholipase D) and degrade (monoacylglycerol lipase and fatty acid amino hydrolase) endocannabinoids may be altered in the hippocampus of AM251 (3 mg/kg)-treated rats fed three different diets: standard diet (normal chow), high-carbohydrate diet (70% carbohydrate) and high-fat diet (60% fat). Results indicated that AM251 reduced caloric intake and body weight gain, and induced a modulation of the expression of ECS-related proteins in the hippocampus of animals exposed to hypercaloric diets. These effects were differentially restricted to either the 2-arachinodoyl glycerol or anandamide signaling pathways, in a diet-dependent manner. AM251-treated rats fed the high-carbohydrate diet showed a reduction of the diacylglycerol lipase alpha : monoacylglycerol lipase ratio, whereas AM251-treated rats fed the high-fat diet showed a decrease of the N-acyl phosphatidylethanolamine phospholipase D : fatty acid amino hydrolase ratio. These results are consistent with the reduced levels of hippocampal endocannabinoids found after food restriction. Regarding the CB1 expression, AM251 induced specific changes focused in the CA1 stratum pyramidale of high-fat-diet-fed rats. These findings indicated that the cannabinoid antagonist AM251 modulates ECS-related proteins in the rat hippocampus in a diet-specific manner. Overall, these results suggest that the hippocampal ECS participates in the physiological adaptations to different caloric diets.

Summary: This study aimed to compare the efficacy and safety of Deg-100 and Gla-300 in adults with type 1 diabetes. The results showed no significant difference in reducing HbA1c between Deg-100 and Gla-300 after 24 weeks. There were no significant differences in HbA1c, nocturnal hypoglycemia, severe hypoglycemia, DQOL, or DTSQ scores between the two groups. However, the incidence rates of hypoglycemia below 3.9mmol/L and 3.0mmol/L were different between the groups and require further confirmation.

Summary: This study describes the sequelae one month after hospital discharge among patients who required ICU admission for severe COVID-19 pneumonia from a multidisciplinary perspective. The main sequelae observed were fatigue, dyspnea, and oxygen therapy on discharge. Rehabilitation plays a key role in improving the quality of life and social integration of these patients.

Summary: This study investigates the potential biomarkers of neurodegeneration in Substance Use Disorder (SUD) patients and finds that the changes in circulating concentrations of Neurofilament Light chain protein (NfL) and Brain-Derived Neurotrophic Factor (BDNF) are associated with cognitive impairment, especially in patients with moderate-severe cognitive dysfunction. Therefore, the combined use of NfL and BDNF (NfL/BDNF ratio) is suggested for monitoring substance-induced neurocognitive disorder.

Acute intrahippocampal administration of melanin-concentrating hormone impairs memory consolidation and decreases the expression of MCHR-1 and TrkB receptors

Summary: Interest in the role of melanin-concentrating hormone (MCH) in memory processes has grown, but studies have provided contradictory findings. This study investigated the effects of MCH on memory consolidation and the BDNF/TrkB system. The results showed that high doses of MCH impaired memory consolidation, but this effect could be reversed by a co-administration with the MCHR-1 antagonist ATC-0175. Additionally, MCH decreased the expression of MCHR-1 and TrkB, linking both endogenous systems to memory processes.

Summary: We conducted a study to explore the potential role of the LPA signaling system as a biomarker in cocaine use disorders (CUD). The study found that patients with CUD had lower plasma concentrations of LPA species and higher mRNA expression of LPA(1) receptor compared to healthy controls. Additionally, a positive association was found between plasma concentration of 20:4-LPA and CUD-related variables. The study also revealed sex differences in LPA concentrations and alterations in rats treated with cocaine, indicating the impact of CUD and sex on LPA signaling.

Summary: Genome-wide association studies (GWAS) are a powerful tool for identifying disease-related biochemical pathways and facilitating drug repurposing. DAGGER is a pipeline used to find approved drugs with repurposing potential by analyzing GWAS and gene expression data. Using this pipeline, we identified midostaurin as a protective drug for Alzheimer's disease and confirmed its effect in both C. elegans and 3xTg AD transgenic mice models. Our pipeline has the potential to be a useful tool for drug repurposing in complex diseases.

The role of PPAR-gamma in memory deficits induced by prenatal and lactation alcohol exposure in mice

Summary: Patients with Fetal Alcohol Spectrum Disorder (FASD) exhibit long-lasting cognitive disabilities, including memory deficits, but the underlying neurobiological mechanisms are not clear. By studying alcohol-exposed mice, researchers found that prenatal and lactation alcohol exposure induces FASD-like memory impairments through the reduction of N-acylethanolamines (NAEs) and peroxisome proliferator-activated receptor gamma (PPAR-gamma) in the hippocampus during a childhood-like period. Pharmacological interventions targeting NAEs and PPAR-gamma were able to improve memory deficits, and overexpression of PPAR-gamma in hippocampal astrocytes mitigated memory impairments induced by alcohol exposure.

Postural orthostatic tachycardia syndrome (POTS), a form of dysautonomia, can occur after COVID-19 infection or vaccination and has debilitating symptoms. This review discusses the etiology, epidemiology, clinical manifestations, diagnosis, treatment, prognosis, and socioeconomic impact of POTS and other dysautonomic disorders after COVID-19 infection and vaccination. It emphasizes the importance of recognizing and managing POTS and informs specialists in various medical fields about the approach to autonomic disorders.

Olive Oil Phenolic Compounds' Activity against Age-Associated Cognitive Decline: Clinical and Experimental Evidence

Summary: Epidemiological studies suggest that consuming olive oil rich in phenolic bioactive compounds may lower the risk of neurodegenerative diseases and improve cognitive performance in the elderly. Preclinical evidence indicates that olive oil phenolic compounds can counteract oxidative stress and neuroinflammation, key processes in neurodegeneration. Clinical evidence suggests that high consumption of olive oil phenolic compounds is associated with better cognitive preservation, independent of dietary patterns. Further research is needed to understand the mechanisms and potential clinical applications.

The study emphasizes the need to control the current use of psychotropic medication as a confounding variable when considering the use of NAEs as biomarkers in SUD.
