Photocytotoxic Oxidovanadium(IV) Complexes of Polypyridyl Ligands Showing DNA-Cleavage Activity in Near-IR Light

Oxidovanadium(IV) complexes [VO(pyphen)(L)]Cl2 (1, 2) and [VO(pydppz)(L)]Cl2 (3, 4), where L is 1,10-phenanthroline (phen in 1 and 3) and dipyrido[3,2-a:2',3'-c]phenazine (dppz in 2 and 4) are prepared and characterized. The crystal structure of [VO(pyphen)(phen)](ClO4)2 (1a) shows a six-coordinate VN5O geometry with a VO2+ moiety in which the polypyridyl ligand binds in a meridional fashion and the phen ligand displays a chelating binding mode with an N-donor site trans to the oxidovanadyl group. The complexes show a dd band within 720-750 nm in DMF. The one-electron paramagnetic complexes are 1:2 electrolytes in DMF. The complexes exhibit an irreversible VIV/VIII redox response near -0.85 V vs. SCE in DMF/0.1 M TBAP. The complexes bind to calf thymus (ct) DNA giving Kb values within 7.5 x 104 to 1.1 x 106 M1. The complexes show poor chemical nuclease activity in the dark and exhibit significant DNA-photocleaving activity in near-IR light of 705 and 785 nm forming .OH radicals. Complexes 2-4 show remarkable photocytotoxicity in HeLa cancer cells. FACS analysis of the HeLa cells treated with complex 4 shows cell death as highlighted by the sub G1 peak. Propidium iodide staining data indicate apoptosis as the primary mode of cell death.

Highly Efficient Ir(III)-Coumarin Photo-Redox Catalyst for Synergetic Multi-Mode Cancer Photo-Therapy

Summary: Four photo-catalysts were synthesized and characterized, showing strong visible light absorption and efficient photo-catalytic oxidation of cellular coenzymes. Among them, Ir2 exhibited superior anti-cancer activity against HeLa cells compared to a clinically used photosensitizer, and demonstrated in vivo photo-induced catalytic anticancer activity in mouse models.

Sonodynamic Therapy with Metal Complexes: A New Promise in Cancer Therapy

Summary: Sonodynamic therapy (SDT) using ultrasound (US) as the drug stimulus has emerged as a potential alternative to photodynamic therapy (PDT) to treat deep-seated tumors with higher tissue penetration capacity. While organic molecules and nanomaterials are commonly used as sonosensitizers in SDT, the potential of metal complexes in this area remains underexplored. Recent literature reports have shown the high potential of metal complexes as sonosensitizers in SDT.

Sonodynamic cancer therapy by novel iridium-gold nanoassemblies

Summary: In this study, a sonosensitizer AuNPs@Ir1 was developed by hybridizing an organometallic Ir(III) complex with ultrasmall gold nanoparticles for efficient tumor sonodynamic therapy. AuNPs@Ir1 rapidly entered cancer cells under ultrasound irradiation and induced cancer cell oncosis. The results demonstrated the excellent in-vitro and in-vivo anticancer profile of AuNPs@Ir1, which has the potential to expand the scope of sonodynamic therapy for metal-based anticancer drugs.

Metastasis suppressor NME1 in exosomes or liposomes conveys motility and migration inhibition in breast cancer model systems

Summary: This study demonstrates that tumor-derived exosomes can suppress metastasis by delivering the NME1 metastasis suppressor to recipient tumor cells, resulting in altered endocytic patterns and reduced cell motility and migration. Proteomic analysis of the exosomes identified differentially expressed proteins that may contribute to these effects. Liposomes containing rNME1 also exhibited similar suppressive effects. These findings expand our understanding of the functions of exosomes and liposomes and provide a novel approach to prevent metastasis.

Platin-C containing nanoparticles: a recipe for the delivery of curcumin-cisplatin combination chemotherapeutics to mitochondria

Summary: Cisplatin, a key player in chemotherapy, has been improved by developing new platinum-based prodrugs with active ligands. Platin-C, a platinum(iv)-based prodrug tethered to curcumin, showed enhanced chemotherapeutic efficacy and targeted mitochondria of cancer cells. It also demonstrated the ability to attenuate cellular inflammation.

A Decentralized Dynamic Pricing Model for Demand Management of Electric Vehicles

Summary: A dynamic pricing model is proposed to manage EV loads locally and mitigate equipment overloading in distribution systems. The model considers the sensitivity of EV owners to battery state of charge and urgency of recharge, and the welfare of the EV fleet operator in terms of net load and electricity price. The proposed method achieves privacy preservation and has shown effectiveness in mitigating transformer overloading during peak load hours.

Photocatalytic glucose-appended bio-compatible Ir(III) anticancer complexes

Summary: Rationally-designed glucose-appended Ir(III) photocatalysts were found to exhibit visible light-induced catalytic NAD(P)H oxidation in aqueous solution. Among them, the highly in vivo biocompatible complex Ir3 showed lysosome and mitochondria targeting necro-apoptotic photo-cytotoxicity against various cancer cell lines and multicellular spheroids while remaining non-toxic in the dark.

Os(ii) complexes for catalytic anticancer therapy: recent update

Summary: In response to the increasing mortality rate associated with cancer and the limitations of Pt-based chemotherapeutics, the concept of catalytic anticancer agents has been introduced, showing promising results with Os(ii) complexes in disrupting essential intracellular processes in cancer cells, ultimately leading to cell death. These complexes have the potential to be novel catalytic cancer drugs with unique anticancer mechanisms.
