The validation and verification of laboratory methods and procedures before their use in clinical testing is essential for providing a safe and useful service to clinicians and patients. This paper outlines the principles of validation and verification in the context of clinical human molecular genetic testing. We describe implementation processes, types of tests and their key validation components, and suggest some relevant statistical approaches that can be used by individual laboratories to ensure that tests are conducted to defined standards. Summary: This study aims to evaluate the association between accuracy estimates, abstract conclusion positivity, and completeness of abstract reporting with acceptance to radiology conferences and journals. The results show that manuscripts with positive abstract conclusions were less likely to be accepted by radiology journals, while there was no significant association between accuracy estimates, abstract conclusion positivity, and completeness of abstract reporting with conference abstract acceptance.

Summary: The study aimed to investigate the prognostic value of different Doppler ultrasound measurements for adverse perinatal outcomes. The results showed that umbilical artery pulsatility index added prognostic value for adverse perinatal outcomes, while other Doppler ultrasound measurements did not further improve prognostic performance.

Summary: Replacing chest X-ray with ultra-low-dose chest CT (ULDCT) in the diagnostic work-up of patients suspected of non-traumatic pulmonary disease at the emergency department has similar effects on short-term functional health, hospital admissions, and length of stay. However, more incidental findings were found in the ULDCT group.

Development of ileal adenomas after ileal pouch-anal anastomosis versus end ileostomy in patients with familial adenomatous polyposis

Arthur S. Aelvoet, Victorine H. Roos, Barbara A. J. Bastiaansen, Roel Hompes, Willem A. Bemelman, Cora M. Aalfs, Patrick M. M. Bossuyt, Evelien Dekker

Summary: This study compared the development of ileal adenomas in patients with familial adenomatous polyposis (FAP) who underwent ileal pouch-anal anastomosis (IPAA) or end ileostomy. The results showed that patients with IPAA had a higher incidence of ileal adenomas compared to those with end ileostomy.

Diagnostic Accuracy of MRI for Solid Renal Masses: A Systematic Review and Meta-analysis

Robert A. Frank, Haben Dawit, Patrick M. M. Bossuyt, Mariska Leeflang, Trevor A. Flood, Rodney H. Breau, Matthew D. F. McInnes, Nicola Schieda

Summary: This article provides a systematic review of the available evidence on the diagnostic accuracy of biparametric (bp)-MRI and multiparametric (mp)-MRI for solid renal masses in differentiating malignant from benign, aggressive from indolent, and clear cell renal cell carcinoma (ccRCC) from other histology. The review reveals deficits in the available evidence, with limited studies and high risk of bias, as well as heterogeneity in definitions, imaging techniques, diagnostic criteria, and outcome measures for solid masses.

Summary: The EU In-Vitro Diagnostic Device Regulation (IVDR) aims to ensure transparent validation of diagnostic devices, traceability of results, and post-market surveillance. Laboratory-developed tests (LDTs) may be used in the absence of suitable commercial devices. The implementation of IVDR may lead to the need for urgent replacement of commercial tests with LDTs, and the workload will depend on various factors.

Experiences of nonpregnant couples after receiving reproductive genetic carrier screening results in Belgium

Eva Van Steijvoort, Hilde Peeters, Hilde Vandecruys, Jasper Verguts, Karen Peeraer, Gert Matthijs, Pascal Borry

Summary: The aim of this study was to assess the satisfaction, anxiety, knowledge retention, psychosocial and counseling-related aspects among couples who chose reproductive genetic carrier screening (RGCS). The findings showed that all participants felt they had enough information to make an informed choice and none of them regretted their decision to have RGCS. The study demonstrated that the information/counseling and reporting strategy used in this context led to high participant satisfaction, increased knowledge, and positive psychosocial and counseling-related outcomes.

Covariate-specific ROC curve analysis can accommodate differences between covariate subgroups in the evaluation of diagnostic accuracy

Jenny Lee, Nick van Es, Toshihiko Takada, Frederikus A. Klok, Geert-Jan Geersing, Jeffrey Blume, Patrick M. Bossuyt, I. P. D. study team IPD study team

Summary: This study applied covariate-specific receiver operating characteristic (ROC) curve analysis to individual patient data on D-dimer testing for excluding pulmonary embolism. The results showed significant differences in D-dimer concentration and performance among age groups, while the distribution of results was similar for sex. Estimating conditional ROC curves can improve the ability to select thresholds with greater applicability.

Few randomized trials in preterm birth prevention meet predefined usefulness criteria

Janneke van t Hooft, Charlotte E. van Dijk, Cathrine Axfors, Zarko Alfirevic, Martijn A. Oudijk, Ben W. . J. Mol, Patrick M. Bossuyt, John P. A. Ioannidis

Summary: By evaluating randomized controlled trials on preterm birth prevention, it was found that only 10% of the trials met more than half of the usefulness criteria, but there was improvement over time. Future research planning should focus on using informative outcomes, patient-centeredness, pragmatism, and transparency.

Summary: This study evaluated the clinical and biological profile of sickle cell anemia children in rural Central Africa. The severity of the disease was found to depend on multiple factors, with fetal hemoglobin being the main modulator of disease severity. These findings can provide valuable information for treatment options.

Summary: Imaging tests are commonly used in healthcare, but concerns about their overuse have overshadowed their benefits in clinical decision-making. This study demonstrates the importance of considering patient-centered outcomes (PCOs) when evaluating the value of imaging tests. The PROD framework categorizes PCOs into four main domains and highlights the interplay between them and the factors that can modify them.
