Summary: This review provides an overview on the current knowledge regarding the adrenal and extra-adrenal effects of ACTH. It focuses on the role of ACTH in adrenal rhythmicity, steroidogenesis, mineralocorticoids, and adrenal growth, as well as its effects on various extra-adrenal organs and tissues. The review suggests re-evaluating the role of ACTH in potential treatment strategies due to its noncanonical effects, which have been overlooked in previous studies.

Summary: X-linked adrenoleukodystrophy (ALD) is a rare inherited neurological disorder that presents challenges for clinical management. This comprehensive review synthesizes current knowledge about the neurocognitive and mental health effects of ALD, providing context for improved patient care. The results highlight predictive factors and brain-behavior relationships associated with the diverse manifestations of ALD, while also identifying gaps for future research.

Summary: This study investigated the HPA-axis response during acute stress in children with Prader-Willi syndrome, and evaluated its changes with age and repeated testing. The results showed that early morning ACTH or 11-DOC levels are not suitable for determining acute stress-related central adrenal insufficiency. Using the 11-DOC peak for test interpretation is less age-dependent than the ACTH peak. Repeated testing of the HPA-axis over time is not required, unless clinically indicated.

Summary: This study retrospectively analyzed data from PAI patients in a tertiary hospital in northern China from 2015 to 2021. The results showed that the causes of PAI in children are diverse and predominantly affect males, with ABCD1 gene defects being one of the most common causes. Whole-exome sequencing is a diagnostic tool, but diagnoses remain unclear in some cases.

Summary: Bilateral adrenal hemorrhage is a rare but important condition that can lead to adrenal insufficiency if not promptly recognized and treated. Diagnosing this condition can be challenging due to its generalized and nonspecific symptoms. We present two cases of patients with acute abdominal pain who were found to have bilateral adrenal hemorrhage through laboratory tests and CT scans, and were subsequently diagnosed with adrenal insufficiency and treated accordingly.

Summary: This case report describes a rare case of adrenal cortical carcinosarcoma in a 78-year-old female patient. The unique histology of this malignancy, with both carcinomatous and sarcomatous components, presents a diagnostic challenge for clinicians. The study emphasizes the importance of considering adrenal carcinosarcomas in the differential diagnosis of retroperitoneal masses.

Summary: The potential for soil carbon sequestration (SCS) in agricultural land varies widely across 24 European countries, with none meeting the aspirational goal of the '4 per 1000' initiative. This highlights the need for a wider range of measures and implementation pathways to achieve the goal. The complexity of SCS measurement approaches and the importance of country-specific knowledge and estimates underscore the urgent need for methodological improvements and standardization in SCS accounting.

Summary: Autoantibodies against the adrenal enzyme 21-hydroxylase are a hallmark manifestation in autoimmune Addison's disease (AAD). This study identified strong associations between copy number variants in the CYP21 region and the risk of AAD, most likely due to linkage disequilibrium with disease-associated HLA class II alleles.

Summary: This study found that Cystatin C (CysC) may act as an effector pathway of glucocorticoids (GCs) by recruiting TREM2+ macrophages, and it may be a potential target for combination cancer immunotherapy.
