Decreased concentration of adiponectin together with a selective reduction of its high molecular weight oligomers is involved in metabolic complications of myotonic dystrophy type 1

Objective: The hormone adiponectin exerts beneficial pleiotropic effects on biological and metabolic processes. Although a well-recognized insulin sensitizer, its characteristic has yet to be clearly defined. Myotonic dystrophy type 1 (DM1) is a rare genetic disorder that features muscle wasting and metabolic comorbidity, and patients have an increased risk of developing type 2 diabetes. We analyzed circulating levels of adiponectin and its oligomers to determine whether their expression correlates with metabolic alterations in DM1 patients. Design and methods: We measured the anthropometric and biochemical features and three insulin resistance (IR) indices (homeostasis model assessment, quantitative insulin sensitivity check index, and McAuley) of 21 DM1 patients and of 82 age-, sex-, and weight-matched controls. In the blood samples of patients and controls, adiponectin levels were measured by ELISA, and its oligomers were characterized by using western blotting and gel filtration. The adiponectin gene was molecularly analyzed in patients. Results: DM1 patients had significantly higher body mass index, waist circumference, triglycerides (TGs), glucose, tumor necrosis factor a, and IR; conversely, they had significantly lower concentrations of total serum adiponectin with a selective, pronounced decrease of its high molecular weight (HMW) oligomers. There was a strong negative correlation between adiponectin and TGs in DM1 patients. Conclusions: Our results endorse the hypothesis that decreased expression of adiponectin together with a selective reduction of its HMW oligomers contributes to the worsening of IR and its metabolic complications in DM1 patients. These findings suggest that adiponectin and HMW oligomers may serve as biomarkers and are promising therapeutic agents for IR and its consequences in DM1.

In Silico Identification of Novel Inhibitors Targeting the Homodimeric Interface of Superoxide Dismutase from the Dental Pathogen Streptococcus mutans

Summary: In this study, a virtual screening method was used to discover a novel inhibitor of the microaerophile Streptococcus mutans' superoxide dismutase (SmSOD), which is responsible for dental plaque formation. The compound ALS-31 was found to inhibit SmSOD and other SODs, affecting their catalytic activity. In addition, ALS-31 was shown to inhibit planktonic growth and biofilm formation of S. mutans cultures. A derivative of ALS-31, ALS-31-9, exhibited improved inhibition power. This research provides insights for future drug design studies to combat dental caries.

Summary: Neuroblastoma is the most common extracranial solid tumor in childhood. The prognosis for patients with metastatic relapse or refractory disease is poor. The integration of genome sequencing into standard clinical practice is necessary for personalized therapy. In this study, whole exome sequencing was performed on two patients with relapsed NB, and actionable genetic variations were identified and treated accordingly. The results demonstrate the feasibility of incorporating clinical WES into pediatric oncology practice.

A Novel Variant in RAD21 in Cornelia De Lange Syndrome Type 4: Case Report and Bioinformatic Analysis

Summary: Cornelia de Lange Syndrome (CdLS) is a rare genetic disorder that affects multiple organs. This study presents clinical and genetic findings of a patient with CdLS type 4, a syndrome that has been rarely described before. The patient exhibited previously reported clinical features of CdLS type 4 as well as additional symptoms not mentioned previously. The novel genetic variant found in this patient suggests its pathogenicity. This report provides valuable insights for counseling future CdLS type 4 cases.

ALK inhibitors may have a significant role in treating high-risk, ALK-mutated NB patients in the era of precision medicine.

A Targeted Mass Spectrometry Approach to Identify Peripheral Changes in Metabolic Pathways of Patients with Alzheimer's Disease

Summary: Alzheimer's disease is the most common cause of dementia in the elderly population. It not only affects the brain, but also disturbs the overall metabolism of the body. Analysis of blood samples from patients with AD revealed significant dysregulation of metabolites, including upregulation of glycerophospholipids and ceramide, and downregulation of glutamic acid, other phospholipids, and sphingolipids. Pathway analysis showed that at least five metabolic pathways related to polar compounds were dysregulated in AD patients.

Label-free liquid biopsy through the identification of tumor cells by machine learning-powered tomographic phase imaging flow cytometry

Summary: In this study, we introduce a machine learning-powered tomographic phase imaging flow cytometry system that can provide high-throughput 3D phase-contrast tomograms of individual cells for the identification of circulating tumor cells in microfluidic cytometry condition. We propose a hierarchical machine learning decision-maker that utilizes 3D morphological features calculated from the cells' refractive index to accurately discriminate tumor cells from white blood cells as well as identify the tumor type. Experimental results demonstrate a success rate of over 97% in identifying tumor cells and an accuracy of over 97% in discriminating between different cancer cell types, highlighting the potential of this stain-free liquid biopsy tool for detecting and classifying circulating tumor cells in blood.

Novel Reversible Inhibitors of Xanthine Oxidase Targeting the Active Site of the Enzyme

Summary: Xanthine oxidase (XO) is a flavoprotein that plays a crucial role in the oxidation of hypoxanthine to uric acid. Abnormal functions of XO can lead to severe diseases, such as gout-causing hyperuricemia and tissue damage. In this study, we identified four non-purine-like compounds (ALS-1, -8, -15, and -28) that directly inhibit XO activity. Among them, ALS-28 showed the highest potency as an inhibitor. These structurally unrelated compounds have the potential to be further developed into lead compounds.
