E-cadherin is a hallmark of epithelial-mesenchymal transition (EMT), which plays a crucial role in cancer metastasis. We previously demonstrated that four and a half LIM protein 2 (FHL2) inhibited E-cadherin expression and promoted invasive potential and EMT in colon cancer. Here, we aim to further define the mechanism underlying the inhibition of E-cadherin by FHL2 in colon cancer. The expression profiles of FHL2 and Snail1 were first observed by Western blot, immunofluorescence and immunohistochemistry. We found that both the protein level and the cellular localisation of Snail1 were quite similar to FHL2 in colon cancer; reciprocal co-immunoprecipitation assay showed that FHL2 was able to bind Snail1 and its intact structure was required. The expression of FHL2 was positively correlated to Snail1 while negatively to E-cadherin and phospho-Snail1. FHL2 over-expression induced the accumulation of Snail1 in the nucleus. Moreover, dual luciferase assay revealed that FHL2 over-expression decreased while FHL2 siRNA increased the transcriptional activities of two E-cadherin promoter constructs which contained E-box sites (Snail1-binding elements). Mutation of E-boxes increased the transcriptional activities and FHL2 expression was involved in the function of mutation. These results suggested that FHL2 negatively regulated E-cadherin transcriptional activity through interaction with Snail1. Summary: Cytokine dynamics in respiratory specimens of COVID-19 patients were studied in this retrospective case-control study. The study found that levels of MPO, ADA, CCL22, TNF alpha, and IL-6 mRNA were significantly higher in infected patients compared to controls, with ADA showing better performance. Additionally, severe COVID-19 patients had lower CCL22 mRNA levels. ADA may be a better biomarker for differentiating between infected and uninfected patients, while CCL22 has potential in stratifying the severity of COVID-19.

Summary: This study examined the effects of SARS-CoV-2 infection and vaccine regimens on vaccine-elicited immunity among adults aged >= 70 years in Hong Kong. The results showed that booster doses enhance immunity in individuals with prior infection, while mRNA vaccine boosters enhance T cell response in non-infected individuals. These findings support the use of booster doses for older adults and the use of mRNA vaccine boosters for non-infected individuals.

Summary: Non-alcoholic fatty liver disease (NAFLD) is a common chronic liver disease that is increasing worldwide, and it is associated with colorectal polyps. This study aimed to investigate the potential of serum microRNAs (miRNAs) in identifying NAFLD in colorectal polyp patients. The results showed that a serum miRNA panel composed of four miRNA pairs significantly identified NAFLD in colorectal polyp patients, and its performance improved when patients with other concurrent metabolic disorders were removed from the analysis. The serum miRNA panel could be a potential diagnostic biomarker for screening NAFLD in colorectal polyp patients for early diagnosis and prevention of disease progression.

Summary: The study aimed to investigate the impact of non-alcoholic fatty liver disease (NAFLD) on the immunogenicity of the BNT162b2 vaccine against wild-type and variant strains of SARS-CoV-2, as well as infection outcome. The results showed that NAFLD patients who received two doses of the BNT162b2 vaccine had good immunogenicity against wild-type and delta variant strains, but poor immunogenicity against the omicron variant, and they were not at a higher risk of infection compared to the control group.

Summary: This study evaluated the evolution of thyroid function and autoimmunity among COVID-19 survivors over 6 months. The results showed that most abnormal thyroid function in acute COVID-19 resolved within 6 months, and SARS-CoV-2 infection did not lead to change in thyroid autoimmunity. Interferon treatment was associated with a modest increase in anti-thyroid antibody titres. Thyroid function and anti-thyroid antibodies did not play a significant role in long COVID.

These findings suggest that molecular profiling could help identify longer survivors in patients with resected PM.
