Purification and characterization of a solvent-stable protease from Geomicrobium sp EMB2

A moderately haloalkaliphilic bacterium, Geomicrobium sp. EMB2, was isolated from the Sambhar Salt Lake located in the western part of India. It secreted an alkaline protease, which was stable and active in the presence of a wide range of organic solvents. The protease was purified by hydrophobic interaction chromatography on Phenyl Sepharose 6 Fast Flow matrix, and a 22.6-fold purification with 51.2% recovery was attained. The apparent molecular mass was estimated to be 38 kDa. The enzyme was stable in the pH range 6.0-12.0, the optimum being 10.0. The Km and Vmax towards casein were found to be 0.10 mM and 526 U/min, respectively. The protease was most active at 50 degrees C. It appeared to be serine type, owing to its sensitivity to phenylmethylsulphonyl fluoride (PMSF). It withstood a range of detergents and surfactants, and exhibited remarkable stability in the presence of solvents having a log P 2. The presence of NaCl or osmolytes exerted a protective effect and further enhanced the stability of the enzyme. These properties make this novel protease potentially useful for catalysis in non-aqueous medium.

A Chemosensor Based on Gold Nanoparticles and Dithiothreitol (DTT) for Acrylamide Electroanalysis

Summary: The study demonstrated the rapid and simple electroanalysis of acrylamide using a gold electrode modified with gold nanoparticles and dithiothreitol, which exhibited enhanced sensitivity and selectivity. Differential pulse voltammetry was used to detect ACR with low limits of detection and quantitation, and a wide linear range. The sensor's estimation of ACR levels in potato chips and coffee samples was consistent with high-performance liquid chromatography results.

Utilizing the β-lactam hydrolyzing activity of β-lactamase produced by Bacillus cereus EMB20 for remediation of β-lactam antibiotics

Computational Exploration of Bio-Degradation Patterns of Various Plastic Types

Summary: Plastic materials are difficult to degrade in the open environment, leading to the infiltration of plastic wastes into natural resources. The current disposal methods are inefficient, and recycling is challenging due to the mixed composition of plastic waste. This research analyzed previously published reports on the biodegradation of different plastic types and found that microbial degradation patterns follow the chemical classification of plastic types. The study suggests potential genera for the bioremediation of mixed plastic waste.

Inhibition and eradication of Pseudomonas aeruginosa biofilms by secondary metabolites of Nocardiopsis lucentensis EMB25

Summary: Biofilm-associated disorders, caused by Pseudomonas aeruginosa, are difficult to treat due to their resistance to the host's immune system and stress factors. In this study, a rare halophilic actinobacteria, Nocardiopsis lucentensis EMB25, was isolated and its secondary metabolites were found to inhibit and eradicate P. aeruginosa biofilm. The secondary metabolites target the quorum sensing pathway and bind to LasR and RhlR. The downregulation of various QS system genes further confirmed the action of secondary metabolites on LasR and RhlR, making actinobacteria a potential source for novel bioactives against biofilm-related infections.
