Effect of in vivo chronic exposure to clotrimazole on zebrafish testis function

Clotrimazole is an azole fungicide used as a human pharmaceutical that is known to inhibit cytochrome P450 (CYP) enzymatic activities, including several steroidogenic CYP. In a previous report, we showed that a 7-day exposure to clotrimazole induced the expression of genes related to steroidogenesis in the testes as a compensatory response, involving the activation of the Fsh/Fshr pathway. In this context, the aim of the present study was to assess the effect of an in vivo 21-day chronic exposure to clotrimazole (30-197 mu g/L) on zebrafish testis function, i.e., spermatogenesis and androgen release. The experimental design combined (1) gene transcript levels measurements along the brain-pituitary-gonad axis, (2) 11-ketotestosterone (11-KT) quantification in the blood, and (3) histology of the testes, including morphometric analysis. The chronic exposure led to an induction of steroidogenesis-related genes and fshr in the testes as well as fsh beta in the pituitary. Moreover, increases of the gonadosomatic index and of the volume proportion of interstitial Leydig cells were observed in clotrimazole-exposed fish. In accordance with these histological observations, the circulating concentration of 11-KT had increased. Morphometric analysis of the testes did not show an effect of clotrimazole on meiotic (spermatocytes) or postmeiotic (spermatids and spermatozoa) stages, but we observed an increase in the number of type A spermatogonia, in agreement with an increase in mRNA levels of piwil1, a specific molecular marker of type A spermatogonia. Our study demonstrated that clotrimazole is able to affect testicular physiology and raised further concern about the impact of clotrimazole on reproduction.

Metabolomics insight into the influence of environmental factors in responses of freshwater biofilms to the model herbicide diuron

Summary: Freshwater biofilms have been increasingly used in ecotoxicology over the past decade to assess environmental stress effects. Untargeted metabolomics is a useful approach for understanding the response of these microbial communities to chemical stress and environmental factors. The study demonstrates the influence of temperature, photoperiod, and flow on the metabolomics response, highlighting the complexity of the relationship between molecular alterations and ecosystem functioning. More research is needed to fully decipher the metabolomics response of freshwater biofilms to chemical stress.

Summary: Precocious male maturation in Atlantic salmon aquaculture leads to reduced welfare and increased production costs. Research on the regulation of follicle-stimulating hormone (Fsh) production in male salmonids revealed up-regulation of gonadotropin-related genes in the pituitaries of maturing males exposed to certain environmental conditions. Ex vivo studies showed the influence of hormones and growth factors on gene expression levels in pituitaries from both immature and maturing males.

Summary: Using germ cell-free (GCF), sterile, dnd-knockout salmon for farming can solve the problems associated with precocious maturation and genetic intmgression of farmed breeds into wild populations. The GCF phenotype of salmon is similar to wild type (WT) counterparts in terms of growth and welfare, with the advantage of never maturing.

A PBPK model to evaluate zebrafish eleutheroembryos' actual exposure: bisphenol A and analogs' (AF, F, and S) case studies

Summary: This study used the zebrafish eleutheroembryo model to assess the toxicity and developmental adverse effects of xenobiotics. A new physiologically based pharmacokinetic (PBPK) model was used to predict the concentration of xenobiotics in the eleutheroembryo tissues. The model was improved by considering factors such as dynamic flows through the embryo, metabolic clearance, and experimental temperature. The model showed good agreement with experimental values and provides a tool for designing new assays and evaluating actual exposure.

Summary: This study applied metabolomics-based approaches to identify toxicity pathways and potential biomarkers in the fish species three-spined stickleback exposed to diclofenac. The results showed significant impacts on the liver metabolome, with upregulation of energy metabolism in females and downregulation in males. The antioxidant system was impacted in males, while the immunity system was down-regulated in females. Several potential metabolic biomarkers were identified.

Role of the Melanocortin System in Gonadal Steroidogenesis of Zebrafish

Summary: Reproduction in both males and females is complex and controlled by various hormonal systems that communicate with the hypothalamic-pituitary-gonadal (HPG) system. This study provides new insights into the effects of melanocortin system on gonadal physiology, highlighting the modulatory role of ACTH and MSH on steroid secretion and the need for further research to fully understand the impact of melanocortins on reproductive function. Our results suggest that melanocortins play a role in regulating vitellogenesis in females and testosterone production in males, indicating a potential endocrine involvement in the reproductive system.

The Piwil1 N domain is required for germ cell survival in Atlantic salmon

Summary: The genetic introgression of farmed salmon into wild populations can harm genetic integrity. To address this issue, researchers have identified the potential role of the Piwil1 protein in germ cell survival in Atlantic salmon. By inducing mutations in Piwil1 using CRISPR-Cas9, they observed a mosaic pattern of mutations in the F0 generation of salmon. These mutants still carried germ cells but exhibited partial germ cell loss and delays in spermatogenesis. Further analysis revealed the importance of the N domain of Piwil1 for its function. By generating F1 fish with different combinations of mutated alleles, they found that complete loss of Piwil1 led to the absence of germ cells, while heterozygous fish displayed an intermediate phenotype with a higher ratio of somatic cells to germ cells.

An end-to-end pipeline based on open source deep learning tools for reliable analysis of complex 3D images of ovaries

Summary: Computational analysis of bio-images using deep learning algorithms has progressed significantly and become more accessible to non-specialists thanks to ready-to-use tools. Recent developments in three-dimensional (3D) imaging of ovaries have also benefited the study of oogenesis mechanisms and female reproductive success. However, analyzing these complex datasets has been a challenge due to the lack of efficient workflows. In this study, we integrated two open-source DL tools, Noise2Void and Cellpose, into an analysis pipeline for 3D follicular content analysis, which showed successful application in different types of ovaries. This pipeline enables automatic and accurate quantification of irregularly stained, low autofluorescence, or heterogeneous follicles sizes in 3D images.
