Metabonomic analysis of serum of workers occupationally exposed to arsenic, cadmium and lead for biomarker research: A preliminary study

Environmental metabonomics is the application of metabonomics to characterize the interactions of organisms with their environment. Metabolic profiling is an exciting addition to the armory of the epidemiologist for the discovery of new disease risk biomarkers and diagnostics. This work is a continuation of research searching for preclinical serum markers in a group of 389 healthy smelter workers exposed to lead, cadmium and arsenic. Changes in the metabolic profiles were studied using Proton Nuclear Magnetic Resonance Spectroscopy on pooled serum samples from both the metal exposed and control groups. These multivariate metabonomic datasets were analyzed with Principal Component Analysis and Partial Least Squares Discriminant Analysis. Analysis of metabolic profiles of people exposed to heavy metals suggests energy metabolism disturbance induced by heavy metals. Changes in lipid fraction (very-low-density lipoprotein - VLDL, low-density lipoprotein - LDL), unsaturated lipids and in the level of amino acids suggest perturbation of the metabolism of lipids and amino acids. This study illustrated the high reliability of NMR-based metabonomic profiling on the study of the biochemical effects induced by the mixture of heavy metals. This approach is capable of identifying intermediate biomarkers of response to toxicants at environmental/occupational concentrations, paving the way to its use in a monitoring of smelter workers exposed to low doses of lead, cadmium and arsenic. Summary: The cosmeceutical field, blending cosmetics and pharmaceuticals, is a highly investigated area where peptides are a group of interesting bioactive ingredients meeting industry requirements. Besides bioactivity, bioavailability and stability have become increasingly important factors when considering peptides as cosmeceutical ingredients. New methods are being developed to enhance peptides' skin-penetration and stability, as evidenced by scientific articles and patents in the cosmeceutical arena.

Monosubstituted Acetophenone Thiosemicarbazones as Potent Inhibitors of Tyrosinase: Synthesis, Inhibitory Studies, and Molecular Docking

Summary: A series of 12 monosubstituted acetophenone thiosemicarbazone derivatives were synthesized and tested for their inhibitory properties against tyrosinase activity, with compound 6 showing the highest effectiveness. All investigated compounds inhibited melanin production in B16F10 cells at the micromolar level. Molecular docking revealed interactions between the sulfur atom of the thiourea moiety and copper ions in the active site.

Phosphinotripeptidic Inhibitors of Leucylaminopeptidases

Metabolomic profiling reveals plasma GlycA and GlycB as a potential biomarkers for treatment efficiency in rheumatoid arthritis

Synthesis of Hybrid Tripeptide Peptidomimetics Containing Dehydroamino Acid and Aminophosphonic Acid in the Chain and Evaluation of Their Activity toward Cathepsin C

Summary: In this study, a new group of hybrid phosphonodehydropeptides composed of glycyl-(Z)-dehydrophenylalanine and structurally variable aminophosphonates were synthesized, and their activity towards cathepsin C was investigated. The results indicate that the inclusion of (Z)-dehydrophenylalanine residue induces ordered conformation. The investigated peptides act as weak or moderate inhibitors of cathepsin C.

Tripeptides conjugated with thiosemicarbazones: new inhibitors of tyrosinase for cosmeceutical use

Summary: The development of skin-care products, including cosmeceuticals containing proven active ingredients, such as peptides, is on the rise. However, the applicability of whitening agents in these products is often limited due to drawbacks like toxicity and lack of stability. In this study, thiosemicarbazone (TSC)-peptide conjugates were found to inhibit diphenolase activity, and were tested as tyrosinase and melanogenesis inhibitors in B16F0 cells. The results showed that TSC1-conjugates had a lower IC50 value than the widely used reference compound kojic acid, making them potential candidates for tyrosinase inhibition.

Lymphoscintigraphic alterations in lower limbs in women with lipedema in comparison to women with overweight/obesity

Summary: This study aims to compare lymphoscintigraphy in women with lipedema and women with overweight/obesity. The results show that lymphatic alterations are present in both groups and are associated with weight and body composition parameters. Therefore, lymphoscintigraphy cannot distinguish between lipedema and overweight/obesity.
