OSM can promote osteoblast differentiation and mineralization, but in vivo administration in adult animals does not significantly increase osteoclast formation due to the negative feedback loop through the osteoclastogenesis inhibitor WNT16.

It can directly kill OBs or suppress osteoclastic functions at low concentrations. Mechanism studies revealed that Procoxacin disrupts the feedback loop of the TGF-β/C-Raf/MAPK pathway and reduces osteogenic and osteolytic lesions in PCa-involved bones, leading to a decrease in metastases. This research provides a drug screening strategy based on the cancer-host microenvironment and demonstrates Procoxacin's potential as a therapeutic agent for PCa bone metastasis.

Summary: Loganic acid extracted from Gentiana lutea L. root extract has shown anti-osteoporotic effects by promoting osteoblastic differentiation and inhibiting osteoclast differentiation. In vivo experiments on ovariectomized mice have demonstrated that loganic acid prevents bone mineral density loss and improves bone structural properties in osteoporotic model mice. These results suggest that loganic acid may be a potential therapeutic candidate for the treatment of osteoporosis.

Summary: Atopic dermatitis (AD) is an inflammatory skin disease caused by a disrupted skin barrier and dominated by type 2 immune responses. Staphylococcus aureus (S. aureus) infection poses a high risk to patients with AD. Interleukin-33 (IL-33) and butyrate have been implicated in the pathophysiology of AD, but their effects on AD aggravated by S. aureus infection are not well understood. In this study, we established an AD-like mouse model with epidermal barrier disruption and examined the effects of S. aureus and butyrate on IL-33 expression and immune response. We found that S. aureus infection exacerbated IL-33 release and skin inflammation, while butyrate attenuated S. aureus-aggravated skin inflammation through histone deacetylase 3 (HDAC3) inhibition. These findings suggest that butyrate may have a positive effect on controlling inflammatory skin conditions in AD aggravated by S. aureus infection.

Prostate-specific antigen (PSA) plays a crucial role in this process by promoting osteomimicry of cancer cells, inhibiting osteoclast differentiation, and facilitating osteoblast proliferation and activation at metastatic sites. Furthermore, PSA activates osteogenic factors while suppressing osteolytic factors, contributing to the predominance of osteoblastic bone metastasis and bone remodeling in prostate cancer.
