Summary: The present study aimed to investigate the interactions of 6-ND with the classical catecholamines, phosphodiesterase (PDE)-3 and PDE4, and protein kinase A in rat isolated atria. The results showed that co-incubation of 6-ND with dopamine, noradrenaline, or adrenaline resulted in significant increases in atrial rate, which was partly due to activation of nerve terminals.

Summary: The study evaluated the effects of a monophasic single electric pulse on the contractility, Ca2+ homeostasis, and recovery of cardiac myocytes. Results showed that electroporation increased intracellular Ca2+ levels immediately, leading to changes in myocyte contraction and Ca2+ homeostasis. Sublethal electric pulses also affected cell contractions and Ca2+ levels, with lethal thresholds depending on cell orientation relative to the electric field.

Summary: Cannabinoids are a potential therapy for chronic pain, but their analgesic efficacy is often not reported in clinical trials. This study found that cannabinoids can produce analgesia by inhibiting voltage gate calcium channels, and different cannabinoids target different types of channels through distinct mechanisms. The cannabinoid CBD binds to and occludes the pore of the Ca(v)3.2 channel, alleviating inflammatory and neuropathic pain.

The interfacial action potentials can lead to local calcium concentration changes, potentially serving as a bioelectrical mechanism for interface sensing.

Summary: The Cav3.2 T-type calcium channel plays a crucial role in various pathological conditions, and its calcium channel function is regulated by phosphorylation by multiple kinases. In this study, we used quantitative mass spectrometry to identify the serine/threonine residues on Cav3.2 that are dephosphorylated by calcineurin. We found six dephosphorylated serine residues in different regions of Cav3.2, and observed that calcineurin binds to and dephosphorylates the C-terminus of Cav3.2. Additionally, we discovered that calcineurin can dephosphorylate a previously known CaMKII-phosphorylated site (S1198) and a novel CaMKII-phosphorylated site (S2137) on Cav3.2. These findings provide insights into the regulation of Cav3.2 through both kinase phosphorylation and calcineurin phosphatase dephosphorylation.

Summary: This study demonstrates that Nobiletin (NOB) has anti-arrhythmic effects by delaying susceptibility to arrhythmias and reducing the incidence of fatal ventricular fibrillation. Whole-cell patch-clamp assays show that NOB reversibly reduces the peak current density of the voltage-gated Na+ channel current and shifts the activation and inactivation curves. Molecular docking analysis reveals the specific interaction between NOB and specific target sites.

Summary: Poly-trans-[(2-carboxyethyl)germasesquioxane] (Ge-132), an organogermanium, is hydrolyzed to 3-(trihydrox- ygermyl)propanoic acid (THGP) in aqueous solutions, and reduces inflammation, pain and cancer, whereas the underlying mechanisms remain unknown. It has been found that THGP can directly trap sulfides and inhibit sulfide-induced enhancement of Cav3.2 activity, leading to pain relief.

By using confocal calcium imaging and patch-clamp techniques, it was found that Pitx2c deletion leads to calcium homeostasis imbalance and abnormal electrical activity, which resemble the abnormalities observed in right atrial myocytes from patients with AF.
