Proteomics study of silver nanoparticles toxicity on Bacillus thuringiensis

Emerging technologies in functional genomics and proteomics provide a way of achieving high-throughput analyses, understanding effects on protein populations and sub-populations and follow up environmental stresses. To accomplish these, the action of homemade spherical Silver nanoparticles colloidal suspension (AgNPs) against Bacillus thuringiensis (isolate from Oryza sativa L rhizosphere) was investigated by a proteomic approach (2-DE and NanoLC/FT-ICR MS identification). Thirty four responsive (up/down regulated) proteins were identified. Proteomic results revealed that an exposure of B. thuringiensis cells with different concentrations of AgNPs resulted in an accumulation of envelope protein precursors, indicative of the dissipation of a proton motive force. Identified proteins are involved in oxidative stress tolerance, metal detoxification, transcription and elongation processes, protein degradation, cytoskeleton remodeling and cell division. The expression pattern of these proteins and their possible involvement in the nontoxicity mechanisms were discussed. (C) 2013 Elsevier Inc. All rights reserved.

Summary: The development of plant gametes is highly sensitive to heatwaves, even lasting for just a few days, adversely affecting pollen maturation and reproductive success. Exposure to heat at 40 degrees Celsius can significantly reduce pollen size after the tetrad stage and decrease cotton boll production by 65%. Interestingly, levels of sugar in pollen grains increase after exposure to heat, ruling out carbohydrate deficits as a likely cause of poor reproductive capacity.

Drug Repurposing and De Novo Drug Discovery of Protein Kinase Inhibitors as New Drugs against Schistosomiasis

Summary: Schistosomiasis is a neglected tropical disease that affects more than 200 million people worldwide. Currently, there is only one drug, praziquantel, available for chemotherapy, but it is ineffective against the larval stages of the parasite and there are concerns about drug resistance. Therefore, it is necessary to develop new drugs targeting different species of schistosomes. Protein kinases have been identified as potential targets, and this article discusses various approaches for finding protein kinase inhibitors for schistosomiasis, including screening natural and synthetic compounds, drug repurposing, and de novo drug development. The current status of kinase inhibitors targeting schistosomal serine/threonine kinases and protein tyrosine kinases is also discussed.
