Intensive use of pesticides, particularly dithiocarbamates, in agriculture often leads to contamination of freshwater ecosystems. To our knowledge, the mechanisms of toxicity to fish by the carbamate fungicide Tattoo that contains mancozeb [ethylenebis(dithiocarbamate)] have not been studied. The present study aimed to evaluate the effects of Tattoo on goldfish gills and blood, tissues that would have close early contact with the pollutant. Exposure of goldfish Carassius auratus to 3,5 or 10 mg L-1 of Tattoo for 96 h resulted in moderate lymphopenia (by 8 percent) with a concomitant increase in both stab (by 66-88 percent) and segmented (by 166 percent) neutrophils. An increase in the content of protein carbonyl groups in blood (by 137-184 percent) together with decreased levels of protein thiols (by 23 percent) and an enhancement of lipid peroxide concentrations (by 29 percent) in gills after exposure to 10 mg L-1 of Tattoo demonstrated the induction of mild oxidative stress in response to Tattoo exposure. At the same time, the activities of selected antioxidant enzymes were enhanced in gills: superoxide dismutase by 18-25 percent and catalase by 27 percent. A 34 percent increment in low molecular mass thiol concentrations (mainly represented by glutathione) also occurred in gills and could be related to increased activity (by 13-30 percent) of glucose-6-phosphate dehydrogenase. The results indicate that Tattoo exposure perturbs free radical processes, i.e. induces mild oxidative stress and enhances the activity of certain antioxidant and associated enzymes in goldfish gills. It is clear that goldfish respond to the presence of waterborne pesticide by adjusting antioxidant defenses through upregulation of activities of antioxidant and associated enzymes. Regulation of the unfolded protein response during dehydration stress in African clawed frogs, Xenopus laevis

Amal Idris Malik, Janet M. Storey, Kenneth B. Storey

Summary: The unfolded protein response (UPR) is involved in dealing with dehydration-induced disruption of protein processing in the tissues of African clawed frogs. Cellular stress markers and activating transcription factors related to UPR were upregulated in response to dehydration stress in frog tissues.

Specific and combined effects of dietary ethanol and arginine on Drosophila melanogaster

Summary: This study investigated the effects of essential amino acid l-arginine and ethanol on the physiological and biochemical parameters of fruit flies. The results showed that excessive l-arginine intake led to various changes in physiology and biochemistry, while ethanol intake increased resistance to starvation but decreased resistance to sodium nitroprusside. The combination of l-arginine and ethanol had interactive effects.

Role of unfolded protein response and ER-associated degradation under freezing, anoxia, and dehydration stresses in the freeze-tolerant wood frogs

Jacques Niles, Gurjit Singh, Kenneth B. Storey

Summary: Wood frogs, the most studied anuran for freeze tolerance, are able to survive in frigid temperatures due to their ability to produce glucose as a natural cryoprotectant. Under freezing, anoxia, and dehydration stresses, the liver and skeletal muscle of wood frogs activate the unfolded protein response (UPR) to enhance protein folding/refolding capacity and stabilize cellular proteins for survival. The ERAD pathway is also involved, with muscle showing preference for proteasomal degradation of misfolded proteins, while liver favors refolding of misfolded/unfolded proteins.

Summary: The marsupial Monito del monte uses torpor to save energy and survive in cold and unpredictable environments. A study found that 14 microRNAs in the heart of Monito del monte showed significant differential expression during torpor. These differentially expressed microRNAs are predicted to regulate pathways related to cardiac function and tissue protection during torpor.

MicroRNA transcriptomics in liver of the freeze-tolerant gray tree frog (Dryophytes versicolor) indicates suppression of energy-expensive pathways

W. Aline Ingelson-Filpula, Hanane Hadj-Moussa, Kenneth B. B. Storey

Summary: The gray tree frog, which can freeze up to 42% of its body water during winter, exhibits rapid and reversible miRNA transcriptional regulation. Through miRNA sequencing analysis using RBiomirGS, we identified 11 miRNAs that are differentially regulated during freezing. These miRNAs play roles in downregulating signaling pathways, apoptosis, and nuclear processes while enhancing ribosomal biogenesis. Overall, miRNA induces energy conservation and prioritizes critical processes for freeze-tolerance survival.

Summary: The mitochondrial genome of the endemic Chinese frog Hyla sanchiangensis was analyzed, and it showed typical gene arrangement and structure. Phylogenetic analysis revealed a close relationship between H. sanchiangensis and Hyla annectans and Hyla tsinlingensis. Positive selection in Hyla and Dryophytes is suggested to be a result of historical cold stress events, but further evidence is required to support this hypothesis.

Role of NADP thorn -dependent isocitrate dehydrogenase from muscle tissue of Rana sylvatica in ROS defense during freeze-tolerance

Amanda M. S. Mattice, Anchal Varma, Kenneth B. Storey

Summary: The wood frog uses freeze tolerance to survive in cold environments. When exposed to freezing temperatures, the frog's body water can freeze, halting vital functions and causing damage. The study found that an enzyme called NADP-dependent isocitrate dehydrogenase (IDH) plays a role in antioxidant defense by maintaining the NADPH pool under stress conditions. The activity of IDH is regulated through reversible phosphorylation, with reduced phosphorylation observed in frozen and dehydrated frogs compared to controls.

Hepatic citrate synthase suppression in the freeze-tolerant wood frog (Rana sylvatica)

Anchal Varma, Kenneth B. Storey

Summary: The regulation of citrate synthase (CS) in the liver of wood frogs during freezing was investigated. Results showed a significant decrease in the activity and catalytic efficiency of CS in the frozen frogs' liver, along with a decrease in phosphorylation levels. These results suggest that CS activity is suppressed and tricarboxylic acid (TCA) cycle flux is inhibited during freezing to support the frogs' survival in harsh winter conditions.

Summary: This study analyzed the cardiac tissue of thirteen-lined ground squirrels during hibernation to identify miRNAs involved in hibernation regulation.
