Biological degradation of cyanobacterial hepatotoxin microcystins in estuarine and coastal water samples from the Patos Lagoon estuarine system, a coastal lagoon situated at the southernmost region of Brazil, was observed. Samples of natural surface water were spiked with purified and semi-purified microcystins (MC-LR and [D-Leu(1)]MC-LR) and their concentrations were monitored by HPLC analysis. After 15 days, the toxins were no longer detectable and after 43 days less than 90% of the initial concentration added to the samples was detected by ELISA. The average degradation rates and the exponential decay rate constants from inside and outside of the estuary were similar. A microcystin degradative bacterium was isolated from the estuarine region. Partial sequence of the 16S rDNA showed a 96% homology with the Burkholderia genus. This genus belongs to the beta subdivision on proteobacteria. This is the first report showing the genus Burkholderia as a cyanobacterial toxin degrader. (c) 2007 Elsevier Inc. All rights reserved.

Development of ELISA Using Recombinant Proteins for the Diagnosis of Mycoplasma hyopneumoniae Infection

Simone Simionatto, Silvana Beutinger Marchioro, Marcelo dos Santos Barbosa, Vanessa Galli, Clarice Brink Brum, Sergio Jorge, Odir Antonio Dellagostin

Summary: This study standardized an ELISA method using six recombinant proteins of Mycoplasma hyopneumoniae for detection of serum antibodies in pigs, showing promising results in both diagnostic tests and seroprevalence surveys in commercial pig herds. The recombinant proteins P95 and P102 were identified as potential targets for future diagnostic tests in differentiating infected animals from vaccinated ones.

Summary: Leptospirosis is an emerging infectious disease caused by pathogenic Leptospira spp. Developing a universal vaccine requires identifying highly conserved epitopes exposed on the bacterial surface. A recombinant BCG vaccine targeting leptospiral TBDRs induced a protective and sterilizing immune response in Syrian hamsters, showing promise for preventing leptospirosis.

Summary: Electrochemical sensors and biosensors are useful for fast, inexpensive, sensitive, and easy detection of various specimens. In this study, a modified electrode was used to detect COVID-19 antibodies, showing a linear selective response. The method has the potential to replace traditional tests due to specific antibody-antigen binding interaction and fast response.

DNA microarray for forensic intelligence purposes: High-density SNP profiles obtained directly from casework-like samples with and without a DNA purification step

Gabriel O. Urtiaga, William B. Domingues, Eliza R. Komninou, Amanda W. S. Martins, Eduardo B. Blodorn, Eduardo N. Dellagostin, Rafael dos S. Woloski, Luciano S. Pinto, Clarice B. Brum, Luciana Tovo-Rodrigues, Vinicius F. Campos

Summary: SNP analyses from a forensic intelligence perspective have proven to be an important tool for limiting the number of suspected offenders and finding missing persons. This study used DNA microarray assay to analyze saliva samples collected from a glass surface using a double swab technique commonly applied at crime scenes. Results showed that reliable genotypes with a confidence level of more than 90% can be obtained with a Call Rate above 69.41%, regardless of the experimental condition. The study suggests that DNA microarray can support criminal investigation teams from a forensic intelligence perspective, and future improvements may eliminate the purification step in crime analysis and investigation.
