Gene Amplification as a Mechanism of Yeast Adaptation to Nonsense Mutations in Release Factor Genes

Evgeniia M. Maksiutenko, Yury A. Barbitoff, Andrew G. Matveenko, Svetlana E. Moskalenko, Galina A. Zhouravleva

Summary: The study revealed that gene amplification is a common mechanism of adaptation to nonsense mutations in release factor genes in yeast. The survival of such mutants may involve feedback control of premature stop codon readthrough, but the exact molecular basis of this feedback control remains unclear.

Differential Interactions of Molecular Chaperones and Yeast Prions

Yury A. Barbitoff, Andrew G. Matveenko, Galina A. Zhouravleva

Summary: Baker's yeast is an important model organism for studying various aspects of eukaryotic cell biology. Yeast prions, self-perpetuating protein aggregates, are used to investigate protein quality control and misfolded proteins. Hsp40 proteins have a central role in prion propagation and other cellular processes such as cell signaling and cytoskeleton also contribute to the interaction with prions in yeast cells.

Processing of Fluorescent Proteins May Prevent Detection of Prion Particles in [PSI+] Cells

Andrew G. Matveenko, Varvara E. Ryzhkova, Natalia A. Zaytseva, Lavrentii G. Danilov, Anastasia S. Mikhailichenko, Yury A. Barbitoff, Galina A. Zhouravleva

Summary: This study reveals that labeling prion aggregates with the red fluorescent protein mCherry may prevent accurate detection of the aggregates. In contrast, fusion with the red fluorescent protein TagRFP-T allows for reliable detection of prion aggregates.

Summary: This article investigates nucleoporins and finds that most of them have potential amyloidogenic properties, but the functional aggregation of nucleoporins is controversial.

How Big Is the Yeast Prion Universe?

Galina A. Zhouravleva, Stanislav A. Bondarev, Nina P. Trubitsina

Summary: The number of yeast prions and prion-like proteins has significantly increased since 1994. Initially, skepticism existed among scientists regarding the use of the term "prion" to describe yeast cytoplasmic elements, but now it is widely accepted that prion-like phenomena are common in yeast. This article provides an overview of yeast prions discovered so far and discusses different approaches used to identify them, as well as the potential for discovering new yeast prions.

Direct proof of the amyloid nature of yeast prions [PSI +] and [PIN +] by the method of immunoprecipitation of native fibrils

Summary: Prions can exist in transmissible states with distinct structures and functions. The Sup35 and Rnq1 proteins in prion state form amyloid fibrils in vivo, as demonstrated by immunoprecipitation and staining with Congo Red dye, providing a new method for identifying pathological and functional amyloids in organisms.
