Ketoconazole modulates the infectivity of Ichthyophonus sp (Mesomycetozoa) in vivo in experimentally injected European sea bass

In vitro studies have confirmed the inhibitory effect of the azol-derivative ketoconazole (KZ) on the growth of Ichthyophonus, an important pathogen causing epizootics in wild and cultured fish. We evaluated the effect of KZ in vivo in European sea bass Dicentrarchus labrax experimentally infected with the same Ichthyophonus isolate. Liposomes were used to vehiculate different doses of KZ to increase the effect on Ichthyophonus and lower the toxicity of the drug, and KZ toxicity was assessed in cultured sea bass juveniles. We also studied the effect of liposome-vehiculated KZ included in medicated food on ichthyophoniasis. KZ causes clear toxic effects in D. labrax juveniles at doses > 80 mg kg(-1), apparent in the reduced survival of fish and histological alterations to livers, kidneys and spleens. Fish injected with Ichthyophonus and treated with KZ dosages of <= 80 mg kg(-1) d(-1) presented lower ichthyophoniasis prevalence, fewer organs infected per fish, and fewer spores in the affected organs than the untreated fish. KZ seems to delay the onset of infection, but cannot stop further progression once established. However, this behaviour is not clearly reflected in the biometric and haematological data collected from these fish. We hypothesise that KZ's delaying effect would increase, if lower infective doses (more similar to natural situations) were used. The drug administration vehicle (liposomes vs. emulsions) did not affect the results. Our data confirm the potential utility of KZ in treating ichthyophoniasis and reveal its low toxicity for sea bass. Nevertheless, the optimal dose and appropriate application protocol remain to be determined.

Diet and Host Genetics Drive the Bacterial and Fungal Intestinal Metatranscriptome of Gilthead Sea Bream

Summary: The present study used a metatranscriptomic approach to investigate the expression of gut microbial genes in farmed gilthead sea bream. The researchers found that fungal transcripts were as abundant as bacterial ones, contrary to previous rRNA amplicon-based studies. The study also revealed that dietary intervention and different growth rates of fish influenced the metatranscriptome, with bacteria primarily shaping vitamin and amino acid metabolism and rhythmic and symbiotic processes, while fungi played a more specific role in configuring host immune, digestive, and endocrine processes.

Summary: This study provides a detailed characterization of sex chromosomes and asexual transmission in Eurasian brine shrimp. It was also found that recombination suppression has independently spread to larger sections of the chromosome in the American and Eurasian lineages. Additionally, the study suggests that parthenogenesis in these shrimp is partly controlled by a locus on the Z chromosome, highlighting the interplay between sex determination and asexuality.

Summary: The AquaIMPACT H2020 EU project aims to improve the competitiveness of European Aquaculture by integrating selective breeding and nutrition, while promoting the production of high-quality fish with limited environmental impact. This study evaluated the changes in gut microbiota with diet along the production cycle in genetically improved fish and reference fish. The results showed that the gut microbiota composition had less influence on the growth of genetically improved fish, but their intestinal function and gene expression profiles were enhanced.

Fish microbiomics: Strengths and limitations of MinION sequencing of gilthead sea bream (Sparus aurata) intestinal microbiota

Summary: This study compared different sequencing platforms for microbial characterization in fish gut mucosal samples. The results showed that MinION is a reliable and accurate tool for assessing intestinal bacteria communities, with good correlation with Illumina results. However, less abundant but biologically important taxa may be masked compared to Illumina sequencing.

A New Parasitic Archamoeba Causing Systemic Granulomatous Disease in Goldfish Extends the Diversity of Pathogenic Endolimax spp.

Summary: A new parasite species, Endolimax carassius, was identified as the causative agent of systemic granulomatous disease in goldfish. Phylogenetic analyses showed that this goldfish parasite species is closely related to Endolimax piscium, the causative agent of systemic granulomatous disease in Senegalese sole. The discovery of a considerable diversity of Endolimax spp. among fish highlights the importance of characterizing these parasites in order to understand their evolution and pathogenic potential.

Sparicotyle chrysophrii experimental infection of gilthead seabream (Sparus aurata): Establishment of an in vivo model reproducing the pathological outcomes of sparicotylosis

Summary: This study explores the best experimental conditions to set up an in vivo infection model capable of mimicking the sparicotylosis signs observed in farmed diseased fish. The experimental setup for parasite transmission consisted of a recipient fish tank with naive GSB receiving water from two S. chrysophrii-infected donor tanks. The study provides an advancement for studying S. chrysophrii's biology and its interaction with its host, as well as for further researching treatment alternatives and prophylactic measures.
