Summary: RELA haploinsufficiency is an autoinflammatory condition characterized by intermittent fevers and mucocutaneous ulcerations. Only eight cases have been reported in the literature, but this study adds four families with three novel and one previously described pathogenic variant in RELA. Clinical features include orogenital ulcers, rash, joint inflammation, and fever. Treatment with glucocorticoids, colchicine, and TNF inhibitors has shown benefit. The study highlights the importance of the NF-kappa B pathway and the regulatory role of RELA in mucosal homeostasis, and suggests considering RELA associated inflammatory disease in patients with Behcet's Disease, particularly those with early onset and/or a strong family history.

Structural modifications and nanotechnology offer potential solutions in enhancing the pharmacokinetic profiles of therapeutic substances.

The mutations result in loss of function of RelA protein, leading to excessive IFN expression and autoimmune response. Summary: In this study, AjI kappa B alpha, a homologue of I kappa B alpha in Japanese eel, was cloned and studied for its regulation of immune responses. The results suggested that AjI kappa B alpha functions as an important negative regulator in the host's innate immunity against bacterial and viral infections by inhibiting the activation of NF-kappa B, AP1, and type I IFN signaling pathways.

Summary: Multiple lines of evidence support the critical role of the NF-kappa B signaling pathway in carcinogenesis, with its activation promoting cell proliferation and inhibiting apoptosis in cancer. Inhibition of NF-kappa B through blocking IKK-alpha/beta enzymes and protecting p65 from acetylation and phosphorylation can lead to tumor suppression. This study utilized a virtual screening approach to identify potential inhibitors for both NF-kappa B and I kappa B proteins, resulting in the identification of three top-ranked compounds for each protein.

Summary: Safe and effective nonsteroidal anti-inflammatory drugs are necessary, and adding amino acids to cultures of microorganisms may increase the likelihood of discovering new secondary metabolites. Two new compounds, myceliostatins A and B, with the myceliothermophin structure were isolated from a methionine-enriched culture of Myceliophthora thermophila. Both compounds exhibited inhibition of LPS-induced NO production and had anti-inflammatory and antioxidant effects by suppressing NF-KB activity and reactive oxygen species production.

Summary: The downregulation of NF-kappa B inhibitor alpha (I kappa B alpha) and subsequent inflammation limit the antitumor potential of MLN4924 in liver cancer. Ectopic expression of I kappa B alpha enhances the antitumor potential of MLN4924. MLN4924 decreases I kappa B alpha through promoting the K48 linkage of ubiquitin to I kappa B alpha. Mechanistic studies show that MLN4924 enhances beta-TrCP protein stability, promoting the ubiquitination of I kappa B alpha and its degradation.

Summary: The study compared the effectiveness of adalimumab plus conventional therapy versus conventional therapy alone in treating patients with retinal vasculitis due to refractory Behcet's uveitis. Results showed that adalimumab plus conventional therapy outperformed conventional therapy alone in improving inflammatory parameters, reducing relapse time, and tapering daily glucocorticoid doses. Adverse events were mainly upper respiratory tract infection and gastrointestinal discomfort in the adalimumab group, while fewer adverse events were observed in the conventional therapy group.
