Summary: Most early stage gastric cancer patients do not have lymph node metastases, but metastases appear in the advanced stage. However, some patients have lymph node metastases in the early stage and none in the advanced stage. To explore the molecular mechanisms involved, clinical samples were collected from patients with early and advanced stage gastric cancer with and without lymph node metastases, as well as metastatic lymph nodes. HOXA11 was identified as a key target that is upregulated in gastric cancer tissues and closely associated with lymphatic metastases. HOXA11 transcriptionally regulates TGF beta 1 expression, activates the TGF beta 1/Smad2 pathway, promotes EMT development, induces VEGF-C secretion, and lymphangiogenesis. These findings provide a plausible mechanism for HOXA11-modulated tumor lymphatic metastasis and suggest HOXA11 as a potential therapeutic target for LN-metastatic gastric cancer.

Summary: Several studies have shown the presence of lymphangiogenesis in metastases of breast cancer, and the number of lymph vessels in metastases is significantly correlated with metastasis size. However, no lymphangiogenesis was detected in tumor-free lymph nodes. D2-40 staining was frequently detected in the sinus endothelium of lymph nodes, and a higher proportion of stained sinus in sentinel lymph nodes was associated with worse overall survival and served as an independent prognostic factor. Further studies are needed to understand the biological and clinical significance of the observed immunophenotypic variations of nodal sinus endothelium.

Summary: This study found that high nuclear expression of transcription factor TEAD4 is associated with lymph node metastasis and high lymphatic vessel density in patients with CRC. YAP/TAZ-TEAD4 complexes transcriptionally upregulated the expression of CCBE1 by directly binding to the enhancer region of CCBE1 in both CRC cells and cancer-associated fibroblasts, which resulted in enhanced VEGFC proteolysis and induced tube formation and migration of human lymphatic endothelial cells in vitro and lymphangiogenesis in a CRC cell-derived xenograft model in vivo. In addition, the bromodomain and extra-terminal domain (BET) inhibitor JQ1 significantly inhibited the transcription of CCBE1, suppressed VEGFC proteolysis, and inhibited tumor lymphangiogenesis in vitro and in vivo.

Summary: The lymphatic system is an important transport system in the human body, playing a crucial role in normal physiology and disease pathogenesis. The role of tumor-associated lymphatic vessels and lymphangiogenesis in lymph node metastasis has been well studied, but the mechanisms and relationship between lymphatic vessels and the dissemination of cancer cells to peripheral organs are still not fully understood. Lymphatic vessels also serve as a communication channel between tumors and the host immune system. Lymphatic endothelial cells have recently been recognized as regulators of tumor immunity and immunotherapy responsiveness.

Summary: Recent progress on the central lymphatic system has revealed the function of perivascular spaces and meningeal lymphatic vessels in clearing senescent astrocytes from the aging brain. Blocking meningeal lymphatic drainage leads to impaired clearance and increased neuroinflammation, while enhancing meningeal lymphatic vessel diameter improves clearance and mitigates neuroinflammation. The findings suggest that meningeal lymphatics could be a therapeutic target for age-related brain disorders.
