The purpose of this study was to explore the role of Treg cells, Th17 cells and cytokines associated with Treg/Th17 differentiation in the occurrence, development and outcome of chronic hepatitis B (CHB). To do so, we detected populations of Treg and Th17 cells and their associated cytokines in the peripheral blood of CHB patients. The populations of Treg cells (CD4(+)CD25(high)CD127(low) T cells) and Th17 cells (CD3(+)CD8(-)IL-17(+) T cells) were analyzed in 46 patients with low to moderate chronic hepatitis B (CHB-LM), 24 patients with severe chronic hepatitis B (CHB-S) and 20 healthy controls (HC) using flow cytometry. The levels of cytokines associated with Treg/Th17 differentiation, including IL-10, TGF-beta 1, IL-17 and IL-23, were measured by enzyme-linked immunosorbent assay (ELISA). Our study showed that the imbalance of Treg and Th17 cells might play an important role in the occurrence, development and outcome of CHB. (c) 2013 Elsevier Inc. All rights reserved.

Effect of liver inflammation on accuracy of FibroScan device in assessing liver fibrosis stage in patients with chronic hepatitis B virus infection

Ling-Ling Huang, Xue-Ping Yu, Ju-Lan Li, Hui-Ming Lin, Na-Ling Kang, Jia-Ji Jiang, Yue-Yong Zhu, Yu-Rui Liu, Da-Wu Zeng

Summary: The study evaluated the diagnostic value of the FibroScan device in assessing liver fibrosis stage in chronic HBV-infected patients and found that liver inflammation affects the accuracy of FibroScan. A non-invasive model was established to predict the risk of misdiagnosis of fibrosis stage using FibroScan.

Scalp Acupuncture Protects Against Neuronal Ferroptosis by Activating The p62-Keap1-Nrf2 Pathway in Rat Models of Intracranial Haemorrhage

Ming-Yue Li, Xiao-Hong Dai, Xue-Ping Yu, Wei Zou, Wei Teng, Peng Liu, Xin-Yang Yu, Qi An, Xin Wen

Summary: The study investigated the prophylactic impact of scalp acupuncture therapy on autologous blood injection murine models of intracerebral hemorrhage (ICH) and found that it could mitigate secondary damage following ICH by moderating ferroptosis, reducing iron sequestration, improving mitochondrial health in nerve cell bodies, and suppressing neuronal ferroptosis. The effects were associated with amplified expression of p62, Nrf2, GPX4 and FTH1, decreased expression of Keap1, and upregulation of FTH1 and GPX4, ultimately leading to enhanced recovery after ICH through the antioxidant pathway and mitigation of lipid peroxidation insults arising from ferroptosis.

Decreased frequency of a novel T-lymphocyte subset, CD3+CD4-CD7+CD57- T cells, in hepatitis B virus-related end-stage liver disease might contribute to disease progression

Xueping Yu, Yijuan Zheng, Dawu Zeng, Yongjun Zhou, Jian Sun, Milong Su, Huatang Zhang, Minhui Zheng, Zhipeng Huang, Wenwu Lin, Richeng Mao, Jiming Zhang, Chunfu Zheng, Zhijun Su

Summary: CD3(+)CD4(-)CD7(+)CD57(-) T cells are decreased in patients with end-stage liver disease, especially those with acute to chronic liver failure, and this decrease is associated with disease severity, prognosis, and complications. IL-22 stimulation promotes apoptosis and reduces the number of CD3(+)CD4(-)CD7(+)CD57(-) T cells. These cells exhibit an immunosuppressive phenotype and low expression levels of cytokines.

Clinical characteristics of patients infected with novel coronavirus wild strain, Delta variant strain and Omicron variant strain in Quanzhou: A real-world study

Huatang Zhang, Wenhuang Chen, Xiaoyi Ye, Yongjun Zhou, Yijuan Zheng, Zhangyan Weng, Jianfeng Xie, Kuicheng Zheng, Zhijun Su, Xibin Zhuang, Xueping Yu

Summary: This study aimed to investigate the clinical features of patients infected with novel coronavirus wild strains, Delta variant strains, and Omicron variant strains. The predominant clinical types were the novel coronavirus wild strain and Delta strain. Patients infected with the Omicron strain were mainly asymptomatic. Significant differences were found in various blood parameters among the three groups. Patients infected with the Omicron strain in Quanzhou presented with mild symptoms and had few comorbidities and a good prognosis, but the negative conversion time of the new coronavirus nucleic acid was still considerably long.

Summary: This study found that the proportion of granulocytic myeloid-derived suppressor cells (gMDSC) increased significantly in patients with Hepatitis B virus-related acute-on-chronic liver failure (HBV-ACLF), and this increase was associated with disease severity, prognosis, and infection. Further experiments revealed that TNF-alpha induced gMDSC expansion and inhibited T cell proliferation and interferon-gamma production through the IDO signaling pathway. These findings have important implications for understanding the pathogenesis of HBV-ACLF and developing potential therapeutic targets.

Current status of recombinase polymerase amplification technologies for the detection of pathogenic microorganisms

Shanshan Zhang, Mingyuan Duan, Shuang Li, Jie Hou, Ting Qin, Zhanwei Teng, Jianhe Hu, Huihui Zhang, Xiaojing Xia

Summary: Rapid detection of pathogenic microorganisms is crucial for epidemiologic identification, prevention, and control in public health. PCR-based methods have limitations in resource-limited areas due to the need for precise temperature control. Recombinase polymerase amplification (RPA) is a new nucleic acid amplification technology that can amplify DNA or RNA at a constant temperature, offering simplicity, high specificity and sensitivity, and short detection time. Microfluidic technology combined with RPA enables rapid detection of pathogenic microorganisms by integrating nucleic acid extraction, amplification, and detection.

microRNA as biomarkers in tuberculosis: a new emerging molecular diagnostic solution

Piyush Agrawal, Aditya Upadhyay, Awanish Kumar

Summary: Tuberculosis is a lethal infectious disease caused by Mycobacterium tuberculosis. During infection, the expression of host microRNAs changes, which could be potential biomarkers for early detection of tuberculosis. Further research is needed to identify potential microRNA biomarkers, but momentum is gaining in this field and microRNAs are expected to become routine approaches for reliable diagnosis and specific therapeutic interventions.
