Gut Microbiota Differs in Composition and Functionality Between Children With Type 1 Diabetes and MODY2 and Healthy Control Subjects: A Case-Control Study

OBJECTIVEType 1 diabetes is associated with compositional differences in gut microbiota. To date, no microbiome studies have been performed in maturity-onset diabetes of the young 2 (MODY2), a monogenic cause of diabetes. Gut microbiota of type 1 diabetes, MODY2, and healthy control subjects was compared.RESEARCH DESIGN AND METHODSThis was a case-control study in 15 children with type 1 diabetes, 15 children with MODY2, and 13 healthy children. Metabolic control and potential factors modifying gut microbiota were controlled. Microbiome composition was determined by 16S rRNA pyrosequencing.RESULTSCompared with healthy control subjects, type 1 diabetes was associated with a significantly lower microbiota diversity, a significantly higher relative abundance of Bacteroides, Ruminococcus, Veillonella, Blautia, and Streptococcus genera, and a lower relative abundance of Bifidobacterium, Roseburia, Faecalibacterium, and Lachnospira. Children with MODY2 showed a significantly higher Prevotella abundance and a lower Ruminococcus and Bacteroides abundance. Proinflammatory cytokines and lipopolysaccharides were increased in type 1 diabetes, and gut permeability (determined by zonulin levels) was significantly increased in type 1 diabetes and MODY2. The PICRUSt analysis found an increment of genes related to lipid and amino acid metabolism, ABC transport, lipopolysaccharide biosynthesis, arachidonic acid metabolism, antigen processing and presentation, and chemokine signaling pathways in type 1 diabetes.CONCLUSIONSGut microbiota in type 1 diabetes differs at taxonomic and functional levels not only in comparison with healthy subjects but fundamentally with regard to a model of nonautoimmune diabetes. Future longitudinal studies should be aimed at evaluating if the modulation of gut microbiota in patients with a high risk of type 1 diabetes could modify the natural history of this autoimmune disease.

Metformin action over gut microbiota is related to weight and glycemic control in gestational diabetes mellitus: A randomized trial

Summary: In women with gestational diabetes mellitus, treatment with metformin led to lower postprandial glycemia and less weight gain compared to treatment with insulin. There were also changes in the composition of gut microbiota, with a decrease in Firmicutes and Peptostreptococcaceae abundance and an increase in Proteobacteria and Enterobacteriaceae abundance in the metformin group. Predicted metabolic pathways related to propionate degradation and ubiquinol biosynthesis were more prevalent in the metformin group. Further studies are needed to understand how these changes impact metabolic control during and after pregnancy.

The Prevotella-to-Bacteroides ratio could serve as a predictive tool for weight loss trajectory. Early after surgery, patients who experienced successful weight loss showed an enrichment in taxa related to beneficial effects on host metabolism.

Summary: This study compared the effects of bexagliflozin tablets and glimepiride on the treatment of patients with type 2 diabetes. It was found that bexagliflozin tablets were noninferior to glimepiride in lowering HbA1c levels, and were superior in reducing body mass and systolic blood pressure. Additionally, bexagliflozin tablets were associated with fewer hypoglycemic events and had a favorable effect on estimated glomerular filtration rate (eGFR).

An overview of vitamins as epidrugs for colorectal cancer prevention

Summary: Gene expression can be regulated by epigenomic modifications such as DNA methylation, histone modification, and chromosome remodeling. Vitamins, as natural bioactive compounds, have been found to modify epigenetic patterns and potentially impact the risk of cancer, including colorectal cancer (CRC). This review examines the relationship between vitamins and CRC risk through epidemiological and observational studies and discusses the potential of using vitamins as epigenetic modifiers for CRC treatment. Although more studies with better designs are needed, understanding the molecular mechanisms of vitamin-mediated epigenetic regulation can help identify effective therapeutic targets for CRC prevention and treatment.

The results demonstrated that melatonin had an inhibitory effect on angiogenesis, and this inhibition was dose-dependent.

Summary: This study suggests that the accumulation of persistent organic pollutants (POPs) in visceral adipose tissue is associated with metabolic syndrome. The levels of certain organic pollutants in adipose tissue were found to be related to indicators of metabolic syndrome such as high blood sugar, high blood pressure, and waist circumference. The study also found that the mixture effect of various organic pollutants was associated with the occurrence of metabolic syndrome.

Summary: This study aimed to analyze the intestinal microbiota of patients with rheumatoid arthritis (RA) and obesity, as well as a higher percentage of fatty tissue. A nested case-control study of 80 RA patients and 80 age and sex-matched controls was conducted, and it was found that obesity was more frequent in RA patients and was associated with differences in gut microbiota. Certain bacterial genera were found to be associated with obesity and fat mass index (FMI) in RA patients.

Summary: In this study, the serum and hepatic levels of branched-chain fatty acids (BCFAs) were evaluated in patients with different stages of nonalcoholic fatty liver disease (NAFLD). The results showed an increase in hepatic BCFAs in NAFLD patients, but no differences were observed in serum BCFAs. Correlation analysis also revealed a relationship between hepatic BCFAs and histopathological diagnosis, as well as other parameters related to NAFLD. Gene expression analysis indicated an upregulation of BCAT1, BCAT2, and BCKDHA mRNA levels in NAFLD patients.
