Summary: Tracing SARS-CoV-2 population transmission chains remains a major challenge, but this study presents an integrated system of genomic surveillance that is capable of detecting infection chains in a large city during ongoing community transmission. The system combines viral surveillance sequencing, genetically based identification of infection clusters, integration of public health authority contact tracing data, and a user-friendly dashboard application. This integrated system can contribute to the monitoring and potentially improve the management of SARS-CoV-2 transmission in the population.

Julia Reinhardt, Virag Sharma, Antigoni Stavridou, Annett Lindner, Susanne Reinhardt, Andreas Petzold, Mathias Lesche, Fabian Rost, Ezio Bonifacio, Anne Eugster

Summary: In this study, resting and antigen-responsive T effector (Tconv) and Treg cells were distinguished using single-cell technologies. Machine learning was used to identify a minimal set of 27 genes that could accurately discriminate the four-cell populations. TRIM was identified as a novel marker to distinguish Treg cells.

Summary: The aim of this study was to develop strategies to identify children from the general population with late-stage presymptomatic type 1 diabetes who may benefit from immune intervention. The study found that screening for islet autoantibodies can detect undiagnosed clinical type 1 diabetes and presymptomatic type 1 diabetes in children, and a novel likelihood ratio score can predict the risk of these children developing clinical diabetes.

Potential of TCR sequencing in graft-versus-host disease

Manisha Goel, Anne Eugster, Johannes Schetelig, Ezio Bonifacio, Martin Bornhaeuser, Cornelia S. Link-Rachner

Summary: Graft-versus-host disease (GvHD) is a common complication following allogeneic haematopoietic stem cell transplantation (allo-HSCT), in which T cells play a key role. Monitoring the changes in the T cell receptor (TCR) repertoire can provide insights into the dynamics of the T cell population and the underlying mechanisms of GvHD.

Summary: This study demonstrates the successful application of an integrated genomic surveillance system in tracing the transmission chains of SARS-CoV-2 and investigating travel-associated infection clusters. The findings show the presence of two genetically defined clusters of SARS-CoV-2 isolates in Dusseldorf, with one cluster linked to nightlife venues and a returning traveler, and the other cluster representing multiple independent introductions followed by community transmission.

Summary: The etiology of type 1 diabetes involves autoimmune responses against pancreatic beta cells. This study found heterogeneity in blood glucose control in children with high genetic risk of type 1 diabetes and suggested that islet autoimmunity occurs concurrently or subsequent to insults on the pancreatic islets.
