Summary: This study identified damaging variants in the FOXI3 gene as a genetic cause of craniofacial microsomia (CFM). Missense variants in the nuclear localization sequence were found to affect the subcellular localization of FOXI3. This study shows that two members of the AP-2 transcription factor family, AP-2 alpha and AP-2ss, are important for normal craniofacial development and signaling. The loss of these transcription factors leads to clefts in the upper face and mandible, as well as fusion of the upper and lower jaws in the hinge region.

Summary: The neural crest (NC) is a multipotent cell population in vertebrate embryos with extraordinary migratory capacity, crucial for vertebrate development. The ongoing role of the Wnt/beta-catenin signalling pathway during NC development, especially in the induction of NC and specification of melanocytes, is discussed. Current understanding shows the continual involvement of Wnt/beta-catenin signalling in activating and maintaining the gene regulatory network during NC induction and pigment cell specification, with implications for emerging models and hypotheses on NC fate restriction.

Summary: Bone morphogenetic protein (BMP) signaling plays a crucial role in the regulation of neural induction, neuronal specification, and neuronal differentiation. However, its role in neural progenitors is still not well understood. By manipulating BMP signaling in zebrafish embryos, researchers discovered that inhibiting BMP signaling during gastrulation can increase the proliferation of neural progenitors and decrease the number of neuronal precursors. This study also revealed that there is a fine balance between BMP and Notch signaling in regulating the proliferation of neural progenitor cells.

This study provides an integrated account of the development of the tuberal hypothalamus.
