Volume Loss of the Medial Temporal Lobe Structures in Subjective Memory Impairment

Background/Aims: Subjective memory impairment (SMI) has been suggested as a manifestation of Alzheimer's Disease (AD) preceding mild cognitive impairment (MCI). In this study, we determined the volumes of the hippocampus, the entorhinal cortex (EC) and the amygdala to provide biological evidence for AD in SMI. Methods: Regional volumetric measures were manually traced on 3-Tesla MRI scans. Results: Total brain volume did not differ between the groups. Individuals with SMI had reduced volumes of the hippocampus bilaterally (right p = 0.001; left p < 0.001), the bilateral EC (right p = 0.031, left p = 0.006) and the right amygdala (p = 0.01) compared to the control group. Conclusion: Volume reduction of bilateral hippocampus, bilateral EC and right amygdala supports the concept of SMI as a very early manifestation of AD prior to MCI. SMI may indicate awareness of a degenerative process that can still be functionally compensated. Copyright (C) 2010 S. Karger AG, Basel

Summary: Recordings from neurons in humans implanted with electrodes reveal the existence of different types of cells related to memory, including those encoding selective and invariant representations of abstract concepts, and memory-selective cells associated with familiarity and episodic retrieval. Insights derived from observing these cells include the activation of semantic representations before episodic representations, the segregation of memory content and memory strength, and the relationship between cell activity and subjective awareness in declarative memory.

Alzheimer's Disease and Its Possible Evolutionary Origin: Hypothesis

Summary: The evolution of the human neocortex enabled us to dominate the planet, but there was a glitch in the development of the entorhinal cortical gateway to the hippocampal memory system, leading to Alzheimer's disease (AD). The aging process causes strain and energy costs on the neurons of the lateral entorhinal cortex, resulting in the formation of toxic amyloid-beta oligomers (A & beta;Os) that initiate AD. This study suggests that early treatment with a positively charged peptide or protein that interacts with A & beta;Os, coupled with an inhibitor of CaSR, may be an effective therapeutic combination for AD.

Spatial memory deficits in Alzheimer's disease and their connection to cognitive maps' formation by place cells and grid cells

Summary: When navigating different contexts, our brain builds a cognitive map to internally represent the territory. Spatial navigation involves complex information processing and integration, with place cells, grid cells, and other neurons forming a neural network critical for self-position and spatial memory. Alzheimer's disease (AD), a neurodegenerative disorder affecting the hippocampus-entorhinal cortex (HP-EC) circuit, results in spatial memory deficits and disorientation. Recent studies suggest impairment in spatial information encoding as a possible mechanism, and virtual reality shows promise for early diagnosis and rehabilitation.

Summary: This study provides evidence that the overexpression of c-Jun N-terminal kinases (JNK) is associated with cognitive impairment in Alzheimer's disease (AD). The results suggest that the overexpression of JNK3 in the EC influences the hippocampus (Hp), leading to cognitive dysfunction and triggering inflammatory signaling and Tau misfolding.

Summary: Research finds that subjective cognitive decline (SCD) in individuals who seek medical help may serve as an initial symptomatic stage 2 of the Alzheimer's disease (AD) continuum. The SCD group shows slightly worse cognition, as well as more subtle functional and behavioral symptoms compared to the control group (CO). It is also discovered that SCD-A+ cases (39.3% of all SCD) exhibit greater hippocampal atrophy, lower cognitive and functional performance, and more behavioral symptoms.

Summary: This study aimed to identify new genes associated with breast cancer risk through whole-exome sequencing. The results revealed 20 novel genes with evidence of association, but larger studies are needed for confirmation. Additionally, known genes such as BRCA1, BRCA2, PALB2, ATM, and CHEK2 were found to be associated with increased risk of breast cancer, although they only explain a fraction of the familial aggregation of the disease.

They found associations between AT(N) framework and protein/lipid hubs, and identified Proprotein Convertase Subtilisin/Kexin Type 7 as a potential causative factor for AD.

Summary: Using data from the HypnoLaus study, we found that the relationship between obstructive sleep apnoea (OSA) and cognitive decline in the elderly population is controversial. Our results showed that lower mean peripheral oxygen saturation during sleep and longer time spent asleep with SpO2 <90% were associated with steeper declines in cognitive scores. This study provides evidence of the contribution of OSA and nocturnal hypoxaemia to cognitive decline in the elderly population.

Epigenome-wide analysis identifies methylome profiles linked to obsessive-compulsive disorder, disease severity, and treatment response

Summary: Obsessive-compulsive disorder (OCD) is a common mental disorder affecting around 2-3% of the population. This study investigated the epigenetic factors associated with OCD by analyzing DNA methylation patterns in 185 OCD patients and 199 controls. The researchers identified differentially methylated CpG positions and validated their findings using a subset of 12 CpGs. These CpGs were found to be related to genes associated with the sweet-compulsive brain hypothesis, suggesting a potential role of aberrant dopaminergic transmission in OCD pathogenesis.
