The performance of an average SUV over a 1-mL-volume sphere within an F-18-FDG-positive lesion resulting in the highest possible value (SUVpeakW) was compared with that of an average SUV computed from the 40 hottest voxels, irrespective of their location within the lesion (SUVmax-40). Methods: Dynamic PET performed in 20 lung cancer lesions yielded for each SUV metric its mean value, relative measurement error, and repeatability (MEr-R). Results: SUVpeakW mean value was significantly 9.66% lower than that of SUVmax-40 (P < 0.0001). SUVpeakW and SUVmax-40 MEr-R were significantly lower than the MEr-R of SUVmax (the hottest voxel): 9.35%-13.21% and 8.84%-12.49% versus 13.86%-19.59%, respectively, (95% confidence limit; P < 0.0001). Although being marginal, SUVpeakW MEr-R was not significantly greater than SUN/(max-40) MEr-R (P = 0.086). Conclusion: SUVmax-40 is more likely to represent the most metabolically active portions of tumors than SUVpeakW, with close variability performance.

Hydrogen Sulfide Metabolism and Pulmonary Hypertension

Lukas Roubenne, Roger Marthan, Bruno Le Grand, Christelle Guibert

Summary: Pulmonary hypertension is a severe disease with no curative treatment available, but hydrogen sulfide (H2S) has been shown to have protective effects against it by regulating hypoxia response and mitochondrial bioenergetics, among other pathways. This suggests that H2S could be a relevant target for new therapeutic strategies.

Summary: Various cell types in pulmonary arteries are constantly exposed to mechanical stimulations under conditions of pulmonary hypertension, with calcium-permeable channels playing a key role in maintaining cellular function and calcium homeostasis. SAC are non-selective cation channels that serve as membrane mechano-transducers in responding to physical forces.

Asthmatic bronchial smooth muscle increases rhinovirus replication within the bronchial epithelium

Summary: This study shows that asthmatic bronchial smooth muscle (BSM) cells enhance the replication of rhinovirus (RV) in bronchial epithelium through increased production of CCL20. Additionally, these asthmatic BSM cells downregulate the antiviral pathway PKR, leading to increased susceptibility of the bronchial epithelium to RV infection.

Cell Confluence Modulates TRPV4 Channel Activity in Response to Hypoxia

Solene Barbeau, Alexandre Joushomme, Yann Chappe, Guillaume Cardouat, Isabelle Baudrimont, Veronique Freund-Michel, Christelle Guibert, Roger Marthan, Patrick Berger, Pierre Vacher, Yann Percherancier, Jean-Francois Quignard, Thomas Ducret

Summary: Cell density is a crucial parameter for the activity of the TRPV4 channel in response to hypoxia. At low density, TRPV4 response to hypoxia is increased, while at high density, TRPV4 response is strongly inhibited.
