Polymorphism in the vascular endothelial growth factor A (VEGFA) gene is associated with serum VEGF-A level and disease activity in rheumatoid arthritis: Differential effect of cigarette smoking

Objective: To assess the impact of common genetic variants in the vascular endothelial growth factor A (VEGFA) gene on circulating VEGF-A levels and disease activity in patients with rheumatoid arthritis (RA). Methods: A cohort (n = 419) of consecutively recruited RA patients of Caucasian origin was studied. Disease activity (DAS28) was recorded on a regular basis (0, 12 and 24 months). Smoking history (never, past and current) was obtained. PCR-RFLP assays were used to determine the genotypes of VEGFA single-nucleotide polymorphisms (SNP) including VEGFA-2578 (rs699947), -460 (rs833061), +405 (rs2010963) and +936 (rs3025039). Circulating levels of VEGF-A were measured in serum samples using a fluorescent bead-based assay system (Luminex (R)). Associations were analyzed using univariate and multivariate statistics. Results: VEGFA-2578 AA genotype was associated with lower serum VEGF-A levels, as was the most frequent haplotype (A(-2578)-C-460-G(+405), 48.1%) within the 5'-flanking region of the gene. The same genotype and haplotype were also associated with decreased disease activity in RA. This was seen only in patients who had never smoked. In multivariate multiple regression models, the VEGFA-2578 SNP was shown to be associated with disease activity at presentation (p = 0.029) and over time (p = 0.016) in patients who never smoked, independent of serum VEGF-A levels and other confounding factors. Conclusion: Genetic variation in the VEGFA gene is associated with serum VEGF-A levels in RA, and shows an association with disease activity in RA patients who have never smoked, independent of serum VEGFA levels. Factors Associated with the Uptake of Genetic Testing for Cancer Risks: A Pathway Analysis Using the Health Information National Trends Survey Data

Summary: Our study aimed to identify pathways from the source of information to the uptake of cancer genetic testing, with consideration of intermediate variables including perceptional, attitudinal, and psychosocial factors. Our findings suggest that only information from particular sources, such as healthcare providers and genetic counselors, had positive and significant effects on people's perceptions of cancer regarding its prevention, detection, and treatment. Attitudinal and psychosocial factors showed much smaller or insignificant effects on the uptake of genetic testing.

Geographical pattern of minerals and its association with health disparities in the USA

Summary: This study aimed to determine common patterns of geographic distribution of health-related minerals in the USA and evaluate their cumulative effects on overall population health. Using county-level data on concentrations of 14 minerals in stream sediments and overall health measurements, latent class analysis identified three clusters of life expectancy-related minerals. The 'infertile' cluster was associated with the shortest life expectancy, highest mortality risks at all ages, and highest mortality rates for various causes. The study provides novel insights into environmental geochemistry and its impact on health disparities in the USA.

Investigating the Marginal and Herd Effects of COVID-19 Vaccination for Reducing Case Fatality Rate: Evidence from the United States between March 2021 to January 2022

Summary: To evaluate the effectiveness of COVID-19 vaccination, it is important to separate the herd effect and marginal effect and parameterize them separately. Through studying the relationship between COVID-19 vaccination coverage and case fatality rate (CFR) at the county level in the U.S., we found three breakpoints where herd effects could potentially exist. Our results highlight the need for public-health researchers to differentiate and quantify the herd and marginal effects when analyzing vaccination data, to inform vaccination strategies and evaluate effectiveness.

Summary: Failure of bone healing after fracture often results in nonunion, but the underlying mechanism of nonunion pathogenesis is poorly understood. This study reveals that the inflammatory microenvironment in atrophic nonunion inhibits the expression of DNA methyltransferases, resulting in overexpression of CtBP1/2. The increased CtBP1/2 acts as transcriptional corepressors that suppress the expression of six genes involved in bone healing. The disruption of this signaling may represent a new therapeutic strategy to prevent nonunion incidence after bone fracture.
