Summary: Peroxisomes play an important role in the regulation of various pathological processes. Pex5 gene is critical for peroxisomal biogenesis and accumulation of reactive oxygen species (ROS). This study found that the deletion of Pex5 in mice led to the formation of multinucleated giant cells, meiotic arrest, delayed DNA repair, abnormal spermatogenesis, and decline in fertility. Loss of Pex5 also impaired peroxisome function, resulting in increased ROS levels, germ cell apoptosis, and decreased autophagy.
