Anti-Diabetic Agents from Natural Products-An Update from 2004 to 2009

Diabetes mellitus (DM), the third killer of the mankind health along with cancer, cardiovascular and cerebrovascular diseases, is one of the most challenging diseases facing health care professionals today. The World Health Organization (WHO) has declared that a DM epidemic is underway. Primary DM and its complications are costly to manage, not only for affected individuals, but also for healthcare systems around the world. Screening of anti-diabetic agents has been extensively investigated in the past decades. Natural products (NPs) have served as a major source of drugs for centuries, and about half of the pharmaceuticals in use today are derived from natural substances. Many natural products especially plants-derived medicines have been recommended for the treatment of DM. The present paper reviews NPs appeared in the literature with potential for DM and also identifies the research needs in this area. It mainly covers the time period from January 2004 to June 2009. The current review is divided into three major sections based on classification of the natural materials involved. The first part focuses on known and some new chemical entities isolated mainly from medicinal plants possessing anti-diabetic properties, including saponins, flavonoids, alkaloids, anthraquinones, terpenes, coumarins, phenolics, polysaccharides, and some other compounds. The second part summarizes crude extract of medicinal plants which are commonly used in the traditional Chinese medical system and have been demonstrated experimental or/and clinical anti-diabetic effectiveness, mainly including Leguminosae, Cucurbitaceae, Araliaceae, Liliaceae, Chenopodiaceae, Solanaceae, Compositae, Campanulaceae, Cornaceae, Rhamnaceae, Scrophulariaceae, Euphorbiaceae, Ginkgoceae, Gramineae, Myrtaceae, Sterculiaceae, Annonaceae, Labiatae, Crassulaceae, and Miscellaneous. The third part lists some compound formulae consisting of extracts of several plants that have been reported as beneficial for the treatment of DM, major involving Xiaokeling tablet, Huang-Lian-Jie-Du-Decoction, Ba-Wei-Di-Huang-Wan and Formula 1.

IQGAP2 was identified as the direct target of ISO in lipid homeostasis regulation, and a novel IQGAP2-CREB-SIRT1 axis was demonstrated for the first time in treating NAFLD/NASH.

Summary: A new ratiometric fluorescent probe NPNA-H2Sn was developed with high sensitivity and advantage of colorimetric imaging for rapid and accurate detection of H2Sn. The probe can be used for evaluating the physiological and pathological functions of H2Sn, providing a powerful molecular tool for research in living biosystems.

Se/Albumin Nanoparticles for Inhibition of Ferroptosis in Tubular Epithelial Cells during Acute Kidney Injury

Summary: This study investigates the role of selenium nanoparticles in alleviating cisplatin-induced acute kidney injury. The results suggest that selenium nanoparticles can reduce renal damage by inhibiting ferroptosis, and our self-developed Se/Albumin nanoparticles show superior therapeutic effects compared to regular selenium nanoparticles.

Norcantharidin promotes cancer radiosensitization through Cullin1 neddylation-mediated CDC6 protein degradation

Summary: The cell division cycle (CDC6) has been identified as a critical molecule contributing to radioresistance in cancer cells. The demethylated form of cantharidin, Norcantharidin (NCTD), has been found to re-sensitize radioresistant cancer cells by inducing CDC6 protein degradation, leading to enhanced cell killing and apoptosis. NCTD promotes CDC6 protein degradation through a Cullin1 neddylation-mediated ubiquitin-proteasome pathway.

A new HClO-activated turn-off mitochondria-targetable NIR fluorescent probe for imaging of osteoarthritis in vivo

Summary: Hypochlorous acid (HClO) is an important biomarker in inflammation, and this study presents a new mitochondria-targetable NIR fluorescent probe, NIR-ClO, for specific analysis and imaging of HClO levels in osteoarthritis. NIR-ClO exhibits high sensitivity and selectivity towards HClO, with a low limit of detection and fast response time, making it a promising tool for future osteoarthritis diagnosis research.

Quantitative profiling of differentially expressed oxylipins in ADSCs under proinflammatory cytokine stimulation

Summary: In this study, the oxylipin profiles of adipose-derived mesenchymal stem cells (ADSCs) under cytokine stimulation were comprehensively analyzed using a UPLC-MS/MS method. The results showed that ADSCs activated by the combination of IFN-gamma and TNF-alpha up-regulated LOX-15 products 7-HDHA and 15-HEPE, which are involved in the pro-resolution phase of inflammation. These findings provide a fundamental understanding of MSC-derived oxylipins and may contribute to optimizing MSC-based therapies.

HSP90β promotes osteoclastogenesis by dual-activation of cholesterol synthesis and NF-κB signaling

Summary: Heat shock protein 90 beta (Hsp90 beta) plays a specific role in osteoclasts by binding to Ikk beta, reducing its degradation, and promoting NF-kappa B signaling. It also increases cholesterol biosynthesis, leading to the expression of osteoclastogenic genes. Inhibiting or deleting Hsp90 beta can alleviate bone loss in osteoporosis.

Targeting treatment of bladder cancer using PTK7 aptamer-gemcitabine conjugate

Summary: In this study, a novel targeting system called protein tyrosine kinase 7 aptamer-Gemcitabine conjugate (PTK7-GEMs) was designed and synthesized to deliver Gemcitabine (GEM) to bladder cancer cells. PTK7-GEMs exhibited specific binding to cancer cells and induced cytotoxicity. In vivo experiments demonstrated that PTK7-GEMs had strong anti-tumor efficacy and excellent biosafety.

CD71/CD44 dual-aptamer-gemcitabine conjugate for tumor co-targeting treatment of bladder cancer

Summary: Conventional chemotherapy cannot specifically target cancer cells, leading to damage to normal tissues and organs. In addition, cancer stem cells (CSCs)-induced tumor regrowth and drug resistance limit the effectiveness of certain chemotherapy drugs. This study developed a CD71/CD44 dual-aptamer-gemcitabine (CD71-CD44-GEMs) conjugate for treating bladder cancer by targeting bladder cancer cells and bladder CSCs. The results showed that CD71-CD44-GEMs had selective binding and high inhibitory effects on bladder cancer both in vitro and in vivo. It also exhibited stronger binding affinity and inhibitory efficacy compared to CD71-GEMs or CD44-GEMs in bladder cancer. Therefore, CD71-CD44-GEMs represents a promising approach for bladder cancer treatment as it can effectively target both bladder cancer cells and CSCs and deliver drugs efficiently.

Summary: This study describes a tailor-made human serum albumin-based selenium nanosystem for the treatment of PD that can overcome the intestinal epithelial barrier and blood-brain barrier. The system has lower toxicity and higher efficacy compared to other selenium species and is able to protect dopamine neurons and improve PD symptoms.

Effects of Starch Addition on KGM Sol's Pasting, Rheological Properties, and Gel Texture

Summary: This study investigated the effects of different types of starch on the texture and flavor of konjac tofu. It was found that adding a certain proportion of corn starch, Canna edulis Ker starch, or potato starch can improve the texture and flavor of konjac tofu, resulting in a better structure and mouthfeel.

Physical, chemical, and rheological properties of konjac glucomannan phosphate ester

Summary: The chemically modified konjac glucomannan phosphate ester (KGMP) has been developed to overcome the limitations of low solubility, instability, and poor fluidity of konjac glucomannan (KGM). The study investigated the physical, chemical, and rheological properties of KGMP with different degrees of substitution (DS). The results show that KGMP has potential applications in temperature-sensitive matrix materials, gelling food, and water-retaining agents.

PFKP is upregulated in 5-fluorouracil-resistant patients and suppresses the antitumor activity of 5-fluorouracil in colorectal cancer in vitro and in vivo

Summary: This study found that overexpression of PFKP can enhance resistance of colorectal cancer to 5-FU by promoting glycolysis. Silencing PFKP, on the other hand, can make colorectal cancer cells more sensitive to 5-FU and increase apoptosis. Therefore, PFKP could be a novel candidate for targeted therapy for 5-FU-resistant colorectal cancer.
