Summary: A nanoparticle system targeted at luteinizing hormone-releasing hormone (LHRH) receptor and responsive to the tumor microenvironment has been developed for selective delivery of chemotherapeutic drugs to triple-negative breast cancer (TNBC) cells. This system shows high drug loading efficiency, receptor-mediated internalization into TNBC cells, and improved cytotoxicity compared to non-targeted counterparts. In vivo studies demonstrate enhanced tumor distribution and penetration of payloads, leading to superior therapeutic efficacy in TNBC models. These results offer new insights into precise drug delivery for TNBC treatment.

Summary: To improve the effectiveness of paclitaxel in treating lung cancer, novel redox-sensitive prodrug nanoparticles were developed, which were surface-decorated with transferrin for targeted delivery. These nanoparticles rapidly released the drug under reducing conditions and exhibited excellent stability, cellular uptake efficiency, and selective toxicity towards cancer cells. In animal models, the transferrin-decorated nanoparticles showed superior tumor-suppressing properties compared to the control nanoparticles and intravenous injection of paclitaxel. Moreover, they demonstrated reduced systemic toxicity. Overall, these transferrin-decorated nanoparticles hold great promise for the intracellular delivery of paclitaxel against metastatic lung cancer.

Summary: In this study, a pH-responsive nanogel was prepared as a potential nanocarrier for CK to enhance its antitumor effects against lung cancer. The nanogel efficiently encapsulated CK and showed high drug loading and encapsulation efficiency. In vitro and in vivo experiments demonstrated the superior inhibitory rates of CK-loaded nanogels against tumor volume compared to CK alone.

Summary: This study investigated core crosslinked micelles based on poly(2-ethyl-2-oxazoline)-block-poly(n-butyl acrylate-co-d,l-homocysteine thiolactone acrylamide) by employing different di- and triamines and nucleophilic ring-opening reaction. The quantity of crosslinked micelle and free block copolymer was quantified by size exclusion chromatography (SEC), and the crosslinking conditions were optimized. Subsequently, micelles crosslinked with cystamine exhibited degradation times of 1.5 h to 5 h in the presence of dithiothreitol (DTT), depending on the composition of the hydrophobic core.

Summary: A novel nanogel composed of poly(acrylic acid) (PAA) and pullulan (Pull) was prepared using a simple and green irradiation protocol. The synthesized nanogels were modified with bovine serum albumin (BSA) and folic acid (FA), and then loaded with doxorubicin (DOX) to obtain a delivery system with tumor-specific targeting ability and enhanced biocompatibility. In vitro studies showed that the DOX release from the nanogels was higher in acidic media, which is advantageous for tumor targeting. MTT assay and DAPI staining demonstrated the effectiveness of DOX-loaded nanogels on cancer cells. Neat nanogels showed no destructive effects on both cell lines. Overall, the biocompatible and tumor-specific nanogels hold promise as safe drug carriers for cancer therapy.

Summary: Doxorubicin and indocyanine green were loaded into silica nanoparticles and wrapped with manganese dioxide to create DOX-IR820@MSN-MnO2 nanoparticles. These nanocomposites exhibit tumor microenvironment response characteristics and excellent photodynamic and photothermal properties. The combination therapy using these nanoparticles significantly reduces cell viability and shows promising applications in cancer treatment.

Summary: In this study, we synthesized trimethyl chitosan-functionalized poly(2-hydroxyethyl methacrylate) (PHEMA-TMC) nanogels via surfactant-free emulsion photopolymerization. TMC, a quaternized derivative of chitosan, was used as the main component along with riboflavin (RF) for redox photo-initiating system. The optimal conditions for synthesis were determined, resulting in cationic PHEMA-TMC nanogels with high monomer conversion and a suitable diameter. Additionally, disulfide-crosslinked PHEMA-TMC nanogels were also prepared, showing redox-induced degradation and release of encapsulated melatonin for potential drug delivery applications.

Summary: A dual pH- and time-dependent polymeric coated capsule was developed for site-specific release of simvastatin in the colon, using nanosuspension and anti-solvent crystallization technique to improve drug solubility. The nanosizing technique transformed the crystalline drug into a more soluble amorphous form, while the coated capsules provided specific delivery of simvastatin in the colon with enhanced cytotoxic effects of the drug nanoparticles against HT29 cells. The findings suggest that the coated capsules using ethyl cellulose and Eudragit S100 polymers could be suitable for colon-targeted delivery of simvastatin.

Summary: This study developed dextran-behenic acid-based nanomicelles for targeted delivery of itraconazole to macrophages to enhance its toxic effects against Leishmania parasites. The optimized formulation showed promising anti-leishmanial activities, suggesting that ITZ-loaded nanomicelles could serve as an alternative therapy for cutaneous leishmaniasis.
